1800MHz电磁波出生前暴露对大鼠海马GFAP、NCAM及GABA受体表达的影响

【摘要】: 目的了解1800MHz电磁波出生前暴露对大鼠海马NCAM和GABA受体表达的影响 方法7周龄SD雌性大鼠32只,雄性16只,适应环境一周后以雌:雄=2:1交配,受孕后雌鼠随机分为4组(两个实验组和两个对照组)。实验组采用1800MHz射频电磁波对大鼠于孕0~20天进行21天连续暴露,每天12小时,功率密度为0.5 mW/cm2和1.0 mW/cm2,各实验组分别设置虚拟暴露作为对照组。暴露期间的温度、湿度、背景噪声等环境条件稳定。暴露完成后在F1代子鼠长至三周龄和七周龄,每组随机抽取8只子鼠采用免疫组织化学方法分别对海马GFAP、NCAM及GABA受体进行染色,然后采用图像分析系统分析海马CA1、CA3和DG区(齿状回)的蛋白表达。 结果1.0mW/cm2暴露组未发现对三周龄、七周龄F1代大鼠海马各区GFAP、NCAM、GABA受体表达有影响,其表达与对照组无统计学意义;0.5mW/cm2暴露组三周龄F1代大鼠海马各区NCAM、GABA受体表达和七周龄GFAP、NCAM、GABA受体表达均出现了下调,其表达与对照组相比有统计学意义。结论本实验条件下,1800MHz功率密度0.5mW/cm2电磁辐射出生前暴露使三周龄大鼠NCAM与GABA受体和七周龄大鼠GFAP、NCAM与GABA受体在海马各区表达下调。
[Abstract]:Objective to investigate the effects of 1800MHz electromagnetic wave exposure before birth on the expression of NCAM and GABA receptors in hippocampus of rats. Methods Thirty-two 7-week-old SD female rats and 16 male rats were used to mate with female: male = 2:1 after adapting to the environment for one week. After conception, female rats were randomly divided into 4 groups (two experimental groups and two control groups). Rats in the experimental group were exposed to 1800MHz radio frequency electromagnetic wave for 21 days, 12 hours a day, and the power density was 0.5 mW/cm2 and 1.0 mW/cm2, respectively. Temperature, humidity, background noise and other environmental conditions are stable during exposure. At the age of 3 weeks and 7 weeks after exposure, 8 offspring from each group were randomly stained with GFAP,NCAM and GABA receptors in hippocampus by immunohistochemical method. Then CA1, in hippocampus was analyzed by image analysis system. Protein expression of CA3 and DG region (dentate gyrus). Results there was no effect on the expression of GFAP,NCAM,GABA receptor in the hippocampus of F1 rats at the age of three weeks and seven weeks in the 1.0mW/cm2 exposure group, but there was no significant difference between the expression of GFAP,NCAM,GABA receptor and that of the control group. The expression of NCAM,GABA receptor and GFAP,NCAM,GABA receptor in hippocampus of three week old F1 generation rats in 0.5mW/cm2 exposure group were down-regulated, and the expression of GFAP,NCAM,GABA receptor was significantly lower than that of control group. Conclusion the expression of NCAM and GABA receptors in three-week-old rats and GFAP,NCAM and GABA receptors in hippocampus of 7-week-old rats were down-regulated by 1800MHz power density 0.5mW/cm2 exposure before birth.
