1型糖尿病患者外周血浆细胞样树突状细胞数量及功能的研究

【摘要】: 目的:分析1型糖尿病患者外周血浆细胞样树突状细胞(PDC)数量及功能变化。 方法:采用流式细胞分析技术检测22例1型糖尿病患者(T1DM)及20例健康人体内PDC细胞的比例及数量;用免疫刺激序列(CpG ODN-2216)刺激外周血单个核细胞(PBMCs)并与PBMCs共培养,通过检测PBMCs产生的IFN-α水平评估PDC的功能。 结果:与健康对照组比较,1型糖尿病患者组外周血PDC的百分比和数量均显著下降(p<0.01);PDC经CpG ODN-2216刺激后产生IFN-α的能力较健康对照组也有显著下降(p<0.05);相关性分析表明,经CpG ODN-2216刺激后IFN-α的产量与PDC数量呈正相关关系(r=0.439,p<0.05)。 结论:1型糖尿病患者体内PDC数量和功能异常,说明PDC可能积极参与了患者机体的免疫紊乱过程;PDC产生的IFN-α减少,可能使患者对感染的控制能力降低而易感。
[Abstract]:Objective: to analyze the changes of (PDC) number and function of plasma cell like dendritic cells (PBMC) in type 1 diabetes mellitus (DM). Methods: the proportion and quantity of PDC cells in 22 patients with type 1 diabetes mellitus (T1DM) and 20 healthy people were detected by flow cytometry. Peripheral blood mononuclear cells (PBMCs) were stimulated by immunostimulatory sequence (CpG ODN-2216) and co-cultured with PBMCs. The function of PDC was evaluated by detecting the level of IFN- 伪 produced by PBMCs. Results: compared with the healthy control group, the percentage and quantity of PDC in peripheral blood of type 1 diabetic patients decreased significantly (p < 0. 01). The ability of PDC stimulated by CpG ODN-2216 to produce IFN- 伪 was significantly lower than that of control group (p < 0. 05). Correlation analysis showed that the yield of IFN- 伪 was positively correlated with the quantity of PDC after CpG ODN-2216 stimulation (r = 0. 439, p < 0. 05). Conclusion: the abnormal number and function of PDC in patients with type 1 diabetes mellitus indicate that PDC may participate in the immune disorder process of the patients, and the decrease of IFN- 伪 produced by PDC may make the patients' ability to control infection lower and susceptible to infection.
