BMP-14与bFGF联合诱导骨髓间充质干细胞向软骨分化的实验研究

【摘要】:目的研究骨形态发生蛋白-14(BMP-14)和碱性成纤维细胞生长因子(bFGF)联合诱导大鼠骨髓间充质干细胞(BMSC)向软骨分化的协同作用。方法采用密度梯度离心法与贴壁分离法相结合分离、纯化大鼠骨髓间充质干细胞,用含10%胎牛血清的L-DMEM培养至第3代作为种子细胞,对照组A组不加生长因子,实验组B、C、D组分别加入100ng/ml BMP-14、10ng/ml bFGF和100ng/ml BMP-14+10ng/ml bFGF,培养24h后各组间行细胞增殖比较,14d后观察各组细胞形态学改变,各组间行Ⅱ型胶原免疫细胞化学染色和阿尔辛蓝染色。结果培养14d后,A、B、C、D4组的Ⅱ型胶原细胞染色阳性率分别为(7.5210±3.7243)%、(40.8271±1.2787)%、(17.2818±1.4242)%、(60.5114±1.7634)%;阿尔辛蓝染结果为:A组阴性,B、C组呈淡染,D组呈蓝色。结论BMP-14和bFGF的联合应用较单个细胞因子可以明显诱导骨髓间充质干细胞向软骨细胞表型分化及促进细胞增殖。
[Abstract]:Objective to study the synergistic effect of bone morphogenetic protein-14 (BMP-14) and basic fibroblast growth factor (bFGF) on the differentiation of rat bone marrow mesenchymal stem cells (BMSC) into cartilage. Methods Rat bone marrow mesenchymal stem cells were purified by density gradient centrifugation and adherent separation. L-DMEM containing 10% fetal bovine serum was used as seed cells for the third passage. After 24 hours of 100ng/ml BMP-14,10ng/ml bFGF and 100ng/ml BMP-14 10ng/ml bFGF, culture, the cell proliferation was compared in the experimental group, and the morphological changes of each group were observed after 14 days.
