黄芪联合生长因子诱导人脐带间充质干细胞分化为神经细胞的实验研究

【摘要】: 目的 研究原代人脐带间充质干细胞(UCMSCs)的生物学特性及其体外向神经细胞诱导分化的条件,为组织化人工神经的制备提供干细胞资源。 方法 本实验中我们通过在无菌条件下取正常足月剖腹产新生儿的脐带,利用组织块培养法获得贴壁细胞并进行传代培养和纯化。取第2代贴壁细胞,免疫组化测定细胞表面CD44、CD105、CD106、CD34、CD45表达情况。取生长状态良好的UCMSCs进行细胞爬片,待细胞生长至60%-70%融合时分为四组进行诱导分化。四组细胞分别加入单纯培养液空白对照组(A组)、单纯生长因子诱导组(B组)、单纯黄芪诱导组(C组)和黄芪联合生长因子诱导液(D组),倒置显微镜下观察其形态并检测其表面神经细胞的标志物如NSE、NF和GFAP的阳性表达率,将结果进行统计学分析。 结果 1.原代培养的组织块在5-6d后可见新生贴壁的细胞,形态类似成纤维细胞,一周后快速增殖,成漩涡状聚集,在生长的过程中呈现间充质干细胞的形态特点,免疫组化检测表面标记CD44、CD105强阳性,CD106弱阳性,CD34、CD45阴性,检测结果和UCMSCs的生物学特征符合。 2.单纯bFGF、单纯黄芪和黄芪联合生长因子都可以将细胞诱导向神经样细胞分化,但是通过诱导后细胞表面神经标志物的测定,黄芪联合生长因子诱导组的细胞NSE、NF和GFAP的阳性表达率也更高,经统计学分析,两者的差异明显,具有统计学意义。 结论 1.中药黄芪联合生长因子可明显促进人UCMSCs分化为神经样细胞,不仅细胞形态发生明显变化,而且从分子生物学水平表现出神经细胞特征。 2.与单纯生长因子诱导组(B组)和单纯黄芪诱导组(C组)相比,黄芪联合生长因子诱导(D组)诱导后的人UCMSCs的NSE、NF和GFAP的阳性表达率也更高,经统计学分析,两者的差异明显,具有统计学意义。
