泡球蚴体外培养模型的建立及细胞亲和性观察

【摘要】: 目的:利用肝癌细胞(Bel-7404)、大肠癌细胞(Caco-2)和宫颈癌细胞(Hela)及其培养上述三种细胞的上清液,在体外对泡球蚴进行培养,建立稳定的体外培养模型,观察泡球蚴的在体外的细胞亲和性及生长、发育过程。方法:培养肝癌细胞(Bel-7404)、大肠癌细胞(Caco-2)和宫颈癌细胞(Hela),留取细胞上清液。取泡球蚴组织,分别与上述三种细胞及含三种细胞培养上清的DMEM完全培养基于37℃5%CO2培养箱中培养38d。实验期间对囊泡进行计数,观察囊泡生长情况,记录囊泡最大、最小直径,绘制生长曲线。培养结束后,取培养囊泡囊液进行蛋白定量。利用SDS-PAGE技术观察囊液中蛋白条带特征。结果:泡球蚴与三种细胞及其上清液中均能共同生长,呈出芽生殖方式,囊泡直径0.5~5.0 mm;囊液蛋白含量分别为:肝癌细胞,3.8mg/ml;肝癌细胞上清液,1.1mg/ml;大肠癌细胞,3.2mg/ml;大肠癌细胞上清液,1.2mg/ml;宫颈癌细胞,2.5mg/ml;宫颈癌细胞上清液,3.8mg/ml。泡球蚴在体外与不同的细胞共培养的初期生长特点基本相似:均在培养初期呈较快速性生长(1~15d);生长中期可见一平台期(15~26d);但后期肝癌细胞及其上清液内培养泡球蚴囊泡保持一个较稳定囊泡数量,而其他结果则表现为逐渐减少。并对肝癌细胞上清液所培养的泡球蚴囊壁DNA经PCR扩增出Em特异性200bp条带。SDS-PAGE观察体内外培养囊液中蛋白在14、47-66kd处有相似条带。结论:1)泡球蚴体外生长因素可不依赖培养细胞本身;2)由于体外与动物体内的培养环境的不同,及不同的共培养细胞可能引起囊泡内囊液的蛋白含量有所差异;3)泡球蚴在体外与肝癌细胞及其上清液的亲和性与其它二种细胞略有不同;4)体内、外培养的囊泡含有相似蛋白;5)利用肝癌细胞、大肠癌细胞和宫颈癌细胞及其上清液建立泡球蚴体外培养模型具有一定的可行性,但可能存在一定的差异性。
