BMP-2、VEGF165双基因共表达质粒在小鼠骨髓基质干细胞的表达

【摘要】: 目的:观察转染BMP-2和VEGF165双基因共表达质粒在小鼠骨髓基质干细胞的表达。方法:脂质体介导下将双基因真核表达质粒pIRES-BMP2-VEGF165导入小鼠骨髓基质干细胞,用RT-PCR和免疫组织化学方法观察BMP-2和VEGF165双基因在小鼠骨髓基质干细胞内的表达。结果:①传代后的mBMSC_S增殖迅速,5-7 d细胞达90%融合。传代后不再以集落方式生长,呈均匀分布生长,细胞形态均匀,与成纤维细胞形态相似,大部分呈长梭形,排列规则。②转染BMP-2、VEGF165的小鼠骨髓基质干细胞有明显的BMP-2和VEGF165 mRNA及其蛋白表达。结论:经转染BMP-2和VEGF165双基因质粒的小鼠骨髓基质干细胞能同时表达以上两种基因。
[Abstract]:Aim: to observe the expression of BMP-2 and VEGF165 co-expression plasmid in mouse bone marrow stem cells (BMSCs). Methods: the double gene eukaryotic expression plasmid pIRES-BMP2-VEGF165 was introduced into mouse bone marrow stroma stem cells mediated by liposomes. The expression of BMP-2 and VEGF165 genes in mouse bone marrow stroma stem cells was observed by RT-PCR and immunohistochemistry. Results: 1 the mBMSC_S proliferated rapidly after passage, and 90% of the cells were fused on the 7th day. After passage, it no longer grows in the form of colony, showing uniform distribution and growth, and the morphology of cells is similar to that of fibroblasts, most of which are long fusiform and regular arrangement. 2 the expression of BMP-2, VEGF165 mRNA and their proteins were obvious in mouse bone marrow stroma stem cells transfected with BMP-2,VEGF165. Conclusion: mouse bone marrow stroma stem cells transfected with BMP-2 and VEGF165 double gene plasmid can express the above two genes at the same time.
