黑加仑籽粉对肾性高血压大鼠血压影响的初步研究

【摘要】:目的:研究黑加仑籽粉对肾性高血压大鼠血压、心脏、肾脏的影响,并探讨其降压机制。方法:制备两肾一夹肾性高血压模型,将造模成功大鼠随机分为模型组、卡托普利组(15mg/kg/d)、牛黄降压片组(0.208g/kg/d)、黑加仑籽粉低剂量组(1.375g/kg/d)、中剂量组(2.75g/kg/d)、高剂量组(5.5g/kg/d)。另取10只大鼠作为假手术组。各组大鼠灌胃给药8周,假手术组、模型组给予等容量蒸馏水。给药前及给药后定期对大鼠血压进行测量,并于末次给药后,腹主动脉采血,测定大鼠血浆中血管紧张素II(AngII)、内皮素(ET)、血栓素B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)含量,血清中超氧化物歧化酶(SOD)活力、一氧化氮(NO)、丙二醛(MDA)含量;取心脏、肾脏测定心脏指数、左心室指数、肾脏指数。结果:给药8周后,测定大鼠血压,与模型组相比,黑加仑籽粉各剂量组均能降低肾性高血压大鼠血压(P0.01);测定大鼠血中因子,与模型组相比,黑加仑籽粉各剂量组ET、TXB2、MDA含量明显降低(P0.01或P0.05),NO含量明显升高(P0.01),黑加仑籽粉中、高剂量组AngII含量明显降低(P0.01或P0.05),黑加仑籽粉高剂量组6-Keto-PGF1α含量、SOD活力明显升高(P0.01或P0.05);测定左心室指数,心脏指数,与模型组相比,黑加仑籽粉各剂量组左心室指数明显降低(P0.01或P0.05),黑加仑籽粉高剂量组心脏指数明显降低(P0.05)。结论:黑加仑籽粉能降低肾性高血压大鼠血压且对其心脏有一定的保护作用趋势,推测黑加仑籽粉的降压途径可能与通过抗氧化作用来保护血管内皮细胞,同时调节血管舒缩因子,改善血管内皮功能有关。
[Abstract]:Aim: to study the effects of blackcurrant seed powder on blood pressure, heart and kidney in renal hypertensive rats and to explore its mechanism of lowering blood pressure. Methods: two kidney and one clip renal hypertension models were established. Successful rats were randomly divided into three groups: captopril group (15 mg / kg / d), Niuhuang Jiangya tablet group (0.208 g / kg / d), blackcurrant seed powder low dose group (1.375 g / kg / d), middle dose group (2.75 g / kg / d), high dose group (5.5 g / kg / d). Another 10 rats were used as sham-operated group. Rats in each group were given equal volume distilled water for 8 weeks, sham operation group and model group. Blood pressure was measured regularly before and after administration of the drug in rats. Blood samples from abdominal aorta were taken after the last administration. The contents of angiotensin IIAngIIA, endothelin (et), thromboxane B 2 (TXB 2) -6-keto PGF 1 伪 in plasma of rats were measured, and the plasma levels of angiotensin II, endothelin (et) and thromboxane B 2 (TXB 2) -prostaglandin F 1 伪 (6-Keto-PGF 1 伪) were measured. The activity of superoxide dismutase (SOD), the content of nitric oxide (no) and malondialdehyde (MDA) in serum, the heart index, left ventricular index and renal index were measured in the heart and kidney. Results: after 8 weeks of administration, the blood pressure of rats was measured. Compared with the model group, the blood pressure of rats with renal hypertension was decreased in each dose group of blackcurrant seed powder, and the blood factor of rats was measured, compared with the model group. The content of TXB2O MDA in black currant seed powder decreased significantly (P 0.01 or P 0.05%), the content of AngII in high dose group significantly decreased P0.01 or P0.05%, and the activity of 6-Keto-PGF1 伪 in high dose group increased P0.01 or P0.05%, and the left ventricular index was measured, and the content of TXB2O in black currant seed powder was significantly increased, and the content of 6-Keto-PGF1 伪 was significantly increased in high dose group of black currant seed powder, and the activity of 6-Keto-PGF1 伪 was significantly increased in the high dose group of blackcurrant seed powder (P 0.01 or P 0.05). Compared with the model group, the left ventricular index of the blackcurrant seed powder group was significantly lower than that of the model group. The left ventricular index of each dose group was significantly lower than that of the model group, and the heart index of the high-dose black currant seed powder group was significantly lower than that of the control group. Conclusion: blackcurrant seed powder can reduce blood pressure and protect heart in renal hypertensive rats. It is suggested that the antihypertensive pathway of blackcurrant seed powder may be related to the protection of vascular endothelial cells by antioxidation. At the same time, regulating vasomotor factor and improving vascular endothelial function are related.
