Trio偶联膜转运机制调节神经元突起生长

神经元突起的生长和导向调节是神经发育生物学的基本问题,而细胞骨架重排和膜运输是神经突起生长和导向的基本过程,研究该过程的调节规律是神经发育生物学的关键课题。大量研究表明,神经细胞可通过多条信号通路分别调节细胞骨架重排和膜运输过程,但这两个过程是否存在偶联调节机制目前还不清楚。本文利用含复杂功能域的Trio分子模型,系统研究了该分子的调节规律、并提出了一个细胞骨架重排和膜运输的偶联机制。Trio分子包含两个鸟苷酸交换因子结构域(GEFD)、一个未知功能的蛋白激酶结构、多个Spectrin结构域等,在体内还存在有4个不同长度的剪切体。我们推测,这些结构的复杂性可能是神经元生长精细调节或协同调节的物质基础。为研究Trio多功能域间的协调调节规律,我们试图对不同结构域、不同剪切体进行系统地功能解析。我们将两个鸟苷酸交换因子结构域GEF1和GEF2分别进行片段敲除,并成功地建立了可遗传的小鼠模型(G1NKO和G2NKO)。遗憾的是,G1NKO小鼠完全不表达Trio蛋白,其表型与Trio全蛋白敲除小鼠完全相同,提示该片段敲除导致了全蛋白敲除,其机制目前还不知晓。有意思的是,GEFD2片段敲除小鼠也...
