沙果渣膳食纤维发酵法制备及其功能性研究

【摘要】:本试验以沙果渣为原料,研究了发酵制备沙果渣膳食纤维的工艺及其产品理化特性和对小鼠的润肠通便作用。主要研究结果如下: 1.首先确定了单菌种发酵试验最佳工艺:米根霉在料液比为1:20,接种量8%,在30℃下发酵3d,SDF得率为12.20%;乳酸杆菌在料液比为1:20,接种量10%,在37℃下发酵20h,SDF得率为10.62%;酵母菌在料液比为1:20,接种量12%,在32℃下发酵24h,SDF得率为9.58%。然后通过混合菌种发酵试验确定了菌种混合的比例为:米根霉:乳酸杆菌:酵母菌=2:2:1。最后经工艺优化确定混合菌种发酵最佳工艺为:料液比1:23、接种量12%、发酵温度32℃、发酵时间3d条件下,SDF得率为13.28%。 2.沙果渣膳食纤维理化特性如下:发酵样品过20目筛的溶胀性在25℃和37℃分别为:4.47、4.53mL/ g,过80目筛的溶胀性在25℃和37℃分别为:4.18、4.32mL/g;过20和80目筛的持水力分别为:6.54、7.32g/g。发酵样品对胆固醇的吸附能力与pH有关,在pH7.0下对胆固醇吸附能力为7.6mg/g,大于pH2.0下的吸附能力5.8mg/g。发酵样品对亚硝酸根离子的吸附能力与pH有关,在pH2.0下对亚硝酸根离子吸附能力为1.17mg/g,大于pH7.0下的吸附能力0.78mg/g。发酵法样品对Cd2+、Pb2+、Cu2+的最大束缚量分别为40.4、39.2、36.5μmol/g。发酵样品的溶胀性、持水力、对胆固醇和亚硝酸根的吸附力以及对重金属离子的束缚量都要好与化学法样品和原粉。 3.对于沙果渣膳食纤维抗氧化活性研究表明:在·O2-体系中,其IC50为8.92mg/mL(Vc的IC50=0.71mg/mL);在·OH体系中,其IC50为4.53mg/mL(Vc的IC50=0.49mg/mL);沙果渣膳食纤维和Vc对DPPH清除低于50%,沙果渣膳食纤维对DPPH清除率最高可达到16.24%,但没有Vc效果好,其最高清除率可达到28.50%。通过将沙果渣膳食纤维加入大豆油来抑制油脂的氧化,同时,也跟其他抗氧化剂的比较进行了研究。 4.通过动物试验,研究了膳食纤维对小鼠排便情况的影响,在造模成功的前提下,与模型组比较中、高剂量组均可增加便秘小鼠肠道墨汁推进率(p0.01)。低、中剂量组小鼠对首次排便时间有显著性影响(P0.05),高剂量组小鼠对首次排便时间有极显著性影响(P0.01);低、中、高剂量组小鼠对首次排黑便时间、8h和12h排便质量有极显著性影响(P0.01)。
