复合益生菌对罗非鱼免疫相关基因的影响

【摘要】:本研究通过把三种复合益生菌添加到饲料中,分别饲喂罗非鱼(Tilapia),确定提升免疫保护率最佳的复合益生菌种类,并从分子水平初步探索复合益生菌对罗非鱼免疫力影响的作用机理。试验共分4组,3个试验组和1个对照组。3个试验组用分别添加了等比例的地衣芽孢杆菌(Bacillus licheniformis)+枯草芽孢杆菌(Bacillus subtilis)、地衣芽孢杆菌(Bacillus licheniformis)+凝结芽孢杆菌(Bacillus coagulans)、地衣芽孢杆菌(Bacillus licheniformis)+粪肠球菌(Enterococcus faecalis)的饲料饲喂罗非鱼,添加量为2‰。饲喂40天,期间每隔10天取罗非鱼肝脏、肾脏、头肾、脾脏以及鳃五个器官,采用实时荧光定量PCR技术检测各器官中环氧化酶-2(COX-2)、白细胞介素-1β(IL-1β)和肿瘤坏死因子-a (TNF-a)这三种免疫相关基因的相对表达量变化情况。40天后,以攻毒试验来检测复合益生菌对罗非鱼抗感染保护率的影响。分别给罗非鱼胸鳍基部注射嗜水气单胞菌、维氏气单胞菌和无乳链球菌三种罗非鱼常见致病菌。试验结果表明:三种不同的复合益生菌都显著(P0.05)诱导罗非鱼五个器官中CoX-2、IL-1β和TNF-a基因的表达,其中地衣枯草组诱导表达效果最明显;基因诱导表达在免疫器官肾脏、头肾和脾脏中要比非免疫器官肝和鳃中明显;地衣枯草组提高了罗非鱼对嗜水气单胞菌(Aeromonas hydrophila)、维氏气单胞菌(Aeromonas veronii)和无乳链球菌(Streptococcus agalactiae)的抗感染保护率;地衣凝结组提高了罗非鱼对嗜水气单胞菌和维氏气单胞菌的抗感染保护率。
