基于分子印迹及鲁米诺化学发光体系检测有机磷农药方法研究

【摘要】:有机磷农药广泛应用于作物的病虫害防治以提高作物的产量和质量,但是其大量使用会使环境遭受污染,并危及人类的健康。因此建立有机磷农药残留的检测方法具有重要的意义。本实验基于分子印迹技术及鲁米诺-过氧化氢化学发光体系,建立了两种有机磷农药残留的检测方法,主要研究结果如下:1.采用本体聚合法,以久效磷为模板分子,甲基丙烯酸为功能单体,乙二醇二甲基丙烯酸酯为交联剂,偶氮二异丁腈为引发剂,合成了久效磷分子印迹聚合物。静态吸附、动态吸附及选择性实验表明所制备的印迹聚合物对久效磷具有较好的选择性和特异吸附性。基于久效磷在碱性介质中能够增强鲁米诺-过氧化氢化学发光反应,以制备的分子印迹聚合物作为吸附剂,建立了一种久效磷的分子印迹固相萃取-流动注射化学发光分析方法。结果显示在最优条件下,化学发光增强值与久效磷浓度在0.0023~10mg/L呈良好的线性关系,最低检出限为0.001 mg/L。对油菜和小白菜分别在三个水平上进行了添加回收实验,两种样品的回收率为89.3%~105.9%。2.以合成的敌百虫-BSA-鲁米诺作为化学发光标记物,本体聚合法合成的敌百虫分子印迹聚合物作为仿生抗体,建立了敌百虫流动注射化学发光仿生免疫分析方法,并对影响竞争性免疫反应的条件进行了优化。结果表明以pH 7.2的PBS作为样品稀释液,过氧化氢浓度为0.4 mol/L,该方法的灵敏度(IC50)为2 mg/L,最低检出限(LOD,IC15)为0.0024 mg/L,本方法对胡萝卜和卷心菜的添加回收率在76.7~89.7%之间。
[Abstract]:Organophosphorus pesticides are widely used in crop pest and disease control to improve crop yield and quality, but their extensive use can cause environmental pollution. Therefore, it is of great significance to establish a method for the detection of organophosphorus pesticide residues. This study was based on molecular imprinting and luminol hydrogen peroxide chemiluminescence system. Two methods for the determination of organophosphorus pesticide residues were established. The main results were as follows: 1. Monocrotophos was used as template molecule, methacrylic acid as functional monomer and ethylene glycol dimethacrylate as crosslinking agent. Monocrotophos molecularly imprinted polymer was synthesized by using azodiisobutyronitrile as initiator. The dynamic adsorption and selective experiments showed that the imprinted polymer had good selectivity and specific adsorbability for monocrotophos, and the monocrotophos could enhance the chemiluminescence reaction of luminol and hydrogen peroxide in alkaline medium. Using the molecularly imprinted polymer as adsorbent, a monocrotophos method for solid-phase extraction and flow injection chemiluminescence analysis of monocrotophos was established. There was a good linear relationship between chemiluminescence enhancement value and monocrotophos concentration in 0.0023~10mg/L. The lowest detection limit was 0.001 mg / L. The experiments of adding and recovering rape and pakchoi at three levels were carried out. The recoveries of the two samples were 89.3% and 105.9%, respectively. Using the synthesized trichlorfon -BSA-Lumino as chemiluminescence marker, the molecularly imprinted polymer of trichlorfon was synthesized by bulk polymerization as bionic antibody. A flow injection chemiluminescent bionic immunoassay for trichlorfon was established, and the conditions affecting the competitive immune reaction were optimized. The results showed that pH 7.2 PBS was used as the sample dilution. The concentration of hydrogen peroxide was 0.4 mol / L, the sensitivity of the method was 2 mg / L and the lowest detection limit was 0.0024 mg / L. The recovery rate of the method for carrot and cabbage was 76.7% and 89.7% respectively.
