不同类茶对高脂模型大鼠减肥和防治非酒精性脂肪肝作用的研究

【摘要】:现代科学研究证明茶叶具有多种保健功效,其中包括降血脂、降血糖、抗氧化、防辐射等,,但不同的茶叶对减肥和预防非酒精性脂肪肝的比较研究鲜见报道。据此,本研究以普洱茶(熟茶)、红茶、乌龙茶(铁观音)、普洱茶(熟茶)茶粉、黑茶茶粉和六堡茶为试验材料,以降脂药物为对照,利用外源性膳食诱导引起实验动物高脂血症及形成非酒精性脂肪肝动物模型之后,采用不同剂量的茶汤对其进行持续30天的灌胃,测定实验大鼠相关肝功能和血清生理生化指标,并进行肝脏组织和腹脂肪病理检测,此外通过分子水平研究,观察靶器官P4502E1和IGF-ImRNA基因的表达情况,判定肝组织病变程度,通过现代医学方法和分子生物学手段分析比较受试茶样对高脂模型大鼠减肥和预防非酒精性脂肪肝的作用,为其保健功效的评价提供基础依据。 研究结果如下: 1、减肥作用 (1)普洱茶(熟茶)、铁观音、红茶、普洱茶(熟茶)茶粉、黑茶茶粉、六堡茶均可有效抑制大鼠体重的增长。 (2)(低、中剂量)普洱茶(熟茶)、(低、高剂量)铁观音、(低、高剂量)红茶、中、低剂量)六堡茶可显著提升胰脂肪酶活性(P<0.01)。 (3)普洱茶(熟茶)、铁观音、红茶、(高剂量)黑茶茶粉和(高剂量)六堡茶促进腹脂肪重量显著降低(P<0.01) (4)通过对脂肪细胞大小测量以及细胞组织显微镜观察结果显示,普洱茶(熟茶)、铁观音、红茶、(中剂量)普洱茶(熟茶)茶粉、(高剂量)、黑茶茶粉(高剂量)六堡茶均可有效抑制肥胖大鼠腹部脂肪细胞的增大,减少大鼠腹部脂肪的过多累积。 2、预防非酒精性脂肪肝 (1)通过对大鼠血清中ALT(谷丙转氨酶)、AST(谷草转氨酶)测定,得出普洱茶(熟茶)、(低、中剂量)铁观音和(低剂量)红茶、普洱茶(熟茶)茶粉、六堡茶、黑茶茶粉可使试验大鼠血清中的AST水平显著降低(P<0.05,P<0.01);普洱茶(熟茶)和(低、高剂量)红茶、(低剂量)普洱茶(熟茶)茶粉可使试验大鼠血清中的ALT水平显著降低(P<0.05,P<0.01)。 (2)通过对大鼠肝匀浆中MDA(丙二醛)、SOD(超氧化物歧化酶)和GSH-Px(谷胱甘肽过氧化物酶)活性测定得出,普洱茶(熟茶)茶粉、黑茶茶粉和六堡茶均可有效改善大鼠肝匀浆组织的氧化损伤。 (3)通过对大鼠肝脏病理组织学观察以及病变评分显示:灌胃不同剂量的普洱茶(熟茶)、六堡茶能够梯度性的改善由高脂饲料引起的肝脏脂肪变性。 3、分子水平研究 (1)与高脂模型组相比,普洱茶(熟茶)、红茶、普洱茶(熟茶)茶粉、各高剂量六堡茶可使大鼠肝脏细胞中P4502E1表达量降低,其中(高剂量)普洱茶(熟茶)、(高剂量)红茶最为显著(P<0.05)。 (2)与高脂模型组相比,普洱茶(熟茶)(高剂量)、铁观音、普洱茶(熟茶)茶粉(高剂量)、黑茶茶粉(中剂量)、六堡茶(中、高剂量)可使肝细胞中IGF-I基因表达量显著升高(P<0.05),其中普洱茶(熟茶)、六堡茶效果最好。 结论:普洱茶(熟茶)、铁观音、红茶、普洱茶(熟茶)茶粉、黑茶茶粉、六堡茶对减肥、非酒精性脂肪肝都具有不同程度的保护作用,但普洱茶(熟茶)、六堡茶的作用更显著。
[Abstract]:Modern scientific research shows that tea has many health benefits, including reducing blood fat, lowering blood sugar, antioxidant, anti radiation, but different tea to lose weight and prevent nonalcoholic fatty liver disease comparative study is rarely reported. Therefore, in this study, Pu'er Tea (tea), red tea, Pu'er, Oolong Tea (Tie Guanyin) tea (tea) tea powder, tea tea powder and tea as experimental material, taking lipid-lowering drugs as control, after the use of exogenous dietary induced animal experimental hyperlipidemia and formation of nonalcoholic fatty liver disease animal model, using different doses of tea for 30 days of intragastric administration on the determination of the the rat liver function and serum biochemical indexes, and pathological examination of liver tissue and abdominal fat, the molecular level study, observe expression of target organ of P4502E1 and IGF-ImRNA genes, determining the degree of liver tissue lesions, through modern The effects of tea samples on weight loss and prevention of non-alcoholic fatty liver disease in high-fat model rats were analyzed and compared by medical methods and molecular biological methods.
The results of the study are as follows:
1, the effect of weight loss
(1) Pu'er Tea (tea), tea, Black Tea, Pu'er Tea (tea) tea powder, tea tea, liupao tea could effectively inhibit the growth of body weight of rats.
(2) (low dose) Pu'er Tea (tea), (low, high dose) Tieguanyin, (low, high dose, low dose) Black Tea, Liupu tea) can significantly enhance the activity of pancreatic lipase (P < 0.01).
(3) Pu'er Tea (tea), tea, Black Tea (Gao Jiliang), black tea and tea powder (high dose) of Liupu tea promote abdominal fat weight significantly decreased (P < 0.01)
(4) through the display of fat cell size and cell and tissue measuring microscope observation results, Pu'er Tea (tea), tea, Black Tea, (middle dose) Pu'er Tea (tea) tea powder, tea powder, tea (Gao Jiliang) (Gao Jiliang) increase of Liupu tea could effectively inhibit the fat rat abdominal fat cells. To reduce abdominal fat in rats with excessive accumulation.
2, prevention of nonalcoholic fatty liver
(1) the rat serum ALT (alanine aminotransferase), AST (aspartate aminotransferase) were obtained Pu'er Tea (tea), (low dose) and Tieguanyin (low dose) Black Tea Pu'er Tea (tea), tea powder, tea, black tea powder can make the blood test in the level of AST was significantly decreased (P < 0.05, P < 0.01); Pu'er Tea (tea) and (low, high dose) Black Tea, (low dose) Pu'er Tea (tea) tea powder can make the serum test in rats of ALT were significantly decreased (P < 0.05, P < 0.01).
(2) the rat liver homogenate MDA (MDA), SOD (superoxide dismutase) and GSH-Px (glutathione peroxidase) activity determined that Pu'er Tea (tea), tea powder, tea tea powder and tea can effectively improve the oxidative injury of liver homogenate tissue of rats.
(3) the score of rat liver lesions and histopathological observation showed that intragastric administration of different doses of Pu'er Tea (tea), can improve the gradient of Liupu tea caused by high fat diet fatty degeneration of the liver.
3. Molecular level studies
(1) compared with model group, Pu'er Tea (tea), Black Tea, Pu'er Tea (tea) tea powder, the high dose of Liupu tea can be decreased P4502E1 expression in liver cells of rats, which (high dose) Pu'er Tea (tea), (Gao Jiliang) was the most significant (P < Black Tea 0.05).
(2) compared with model group, Pu'er Tea (tea), tea (Gao Jiliang), Pu'er Tea (tea) tea powder (Gao Jiliang), black tea powder (middle dose), Liupu tea (in high doses) can make the IGF-I gene expression in liver cells increased significantly (P < 0.05) among them, Pu'er Tea (tea), the best effect of liupao tea.
Conclusion: Pu'er Tea (tea), tea, Black Tea, Pu'er Tea (tea) tea powder, tea tea, liupao tea to lose weight, all have different degrees of protection of nonalcoholic fatty liver disease, but Pu'er Tea (tea), significantly more Liupu tea role.
