【摘要】: [目的]研究转化生长因子-β1(transforming growth factor-β1,TGF-β1)和碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)在不同胎龄胎儿角膜的表达及两种因子间表达的差异,了解TGF-β1和bFGF在正常角膜胚胎发育过程中的作用及变化规律,为角膜的胚胎发育提供相关资料。 [方法]收集11~20周各种方法引产的胎儿23例共46只眼,采用免疫组织化学链酶亲和素—生物素—过氧化物酶复合物(streptavdin-biotin-complex,SABC)的方法,检测各胎龄胎儿角膜中TGF-β1和bFGF的表达,并分析TGF-β1与bFGF的相关性。 [结果]1、TGF-β1和bFGF在各胎龄角膜上皮层均有表达。 2、TGF-β1和bFGF在各胎龄角膜其它层次内均未见表达。 3、TGF-β1和bFGF在角膜上皮层的表达均随胎龄的增加而减少,两者比较差异具有统计学意义(P<0.05)。 4、相同胎龄胎儿角膜上皮层内TGF-β1与bFGF的表达有显著性差异,TGF-β1的表达水平高于bFGF的表达水平,两者比较差异具有统计学意义(P<0.05)。 5、经Pearson相关性分析:TGF-β1与bFGF呈正相关(r=0.8037),具有统计学意义(P<0.05)。 [结论]1、各个胎龄的胎儿角膜上皮层均有TGF-β1和bFGF的表达,说明二者均在胎儿角膜上皮发育中发挥一定的作用。 2、各个胎龄的胎儿角膜其它层次内未见TGF-β1和bFGF的表达,具体机制不清。 3、TGF-β1和bFGF在胎儿角膜上皮层的表达均随胎龄的增加而减少。 4、相同胎龄胎儿角膜上皮层内TGF-β1的表达高于bFGF的表达。 5、TGF-β1和bFGF呈正相关,说明TGF-β1和bFGF在胎儿角膜上皮的发育中有协同作用。
[Abstract]:[Objective] Study on transforming growth factor beta 1 (transforming growth factor- TGF- beta 1, beta 1) and basic fibroblast growth factor (basic fibroblast, growth factor, bFGF) differential expression expression in fetal cornea and two kinds of factors, to understand the process in normal cornea and the role in embryonic development the regularity of TGF- beta 1 and bFGF, to provide relevant information for corneal embryo development.
[Methods] from 11 to 20 weeks of induction of various methods of fetal 23 cases of 46 eyes, using immunohistochemical streptavidin - biotin - peroxidase complex (streptavdin-biotin-complex, SABC) method, the expression of the fetal cornea in TGF- beta 1 and bFGF, and correlation analysis of TGF- beta 1 with bFGF.
[results]1, TGF- beta 1 and bFGF were expressed in the corneal epithelium at all fetal age.
2, TGF- beta 1 and bFGF were not expressed in the other layers of the cornea of all gestational ages.
3, the expression of TGF- beta 1 and bFGF in the corneal epithelium decreased with the increase of fetal age, and the difference was statistically significant (P < 0.05).
4, there was a significant difference in the expression of TGF- beta 1 and bFGF in the corneal epithelium of the same gestational age. The expression level of TGF- beta 1 was higher than that of bFGF, and the difference between them was statistically significant (P < 0.05).
5, after Pearson correlation analysis, TGF- beta 1 was positively correlated with bFGF (r=0.8037), and was statistically significant (P < 0.05).
[conclusion]1, the expression of TGF- beta 1 and bFGF in all fetal corneal epithelium at all fetal age shows that all two of them play a role in the development of fetal corneal epithelium.
2, there is no expression of TGF- beta 1 and bFGF in the other layers of fetal cornea at all fetal age, and the specific mechanism is not clear.
3, the expression of TGF- beta 1 and bFGF in the epithelial layer of fetal cornea decreased with the increase of fetal age.
4, the expression of TGF- beta 1 in the corneal epithelium of the fetus of the same gestational age was higher than that of bFGF.
5, there is a positive correlation between TGF- beta 1 and bFGF, indicating that TGF- beta 1 and bFGF have synergistic effects in the development of fetal corneal epithelium.
