【摘要】：目的:本研究旨在探讨脐血间充质干细胞对T淋巴细胞亚群及其细胞因子分泌的影响。方法:采用密度梯度离心法分离培养扩增的脐血间充质干细胞,应用流式细胞术检测脐血间充质干细胞与淋巴细胞混合培养后T淋巴细胞亚群及其细胞因子IL-4和IFN-γ的变化情况。结果:T淋巴细胞与脐血间充质干细胞混合培养后,淋巴细胞增殖受到抑制,CD4+T细胞亚群增加,CD8+T细胞亚群减少;根据细胞因子的变化,Th1和Tc1比率较对照组有显著降低,而Th2和Tc2的比率却有轻度上升。结论:脐血间充质干细胞能够抑制T淋巴细胞尤其是CD8+T细胞的增殖。此外,它还能降低与T淋巴细胞共培养体系中的Th1和Tc1细胞,升高Th2和Tc2细胞。脐血MSCs具有临床应用潜力,可望预防和治疗GVHD。
[Abstract]:Objective: This study aimed to investigate the effect of umbilical cord blood mesenchymal stem cells on T lymphocyte subsets and cytokines. Methods: isolated cultured mesenchymal stem cells from umbilical cord blood by density gradient centrifugation and the detection of changes of umbilical cord blood mesenchymal stem cells mixed with lymph cell culture after T lymphocyte subsets and cytokines IL-4 and IFN- by using flow cytometry. Results: T cells and human umbilical cord blood mesenchymal stem cells after co cultured lymphocytes proliferation was inhibited, CD4+T cell subsets increased, decreased CD8+T cell subsets; cytokines according to the changes of Th1 and Tc1 ratio was significantly lower than the control group, and the ratio of Th2 and Tc2 has slightly increased. Conclusion: umbilical cord blood mesenchymal stem cells can inhibit T lymphocytes especially CD8+T cell proliferation. In addition, it can also reduce and T lymphocytes co cultured Th1 and Tc1 cells, The increase of Th2 and Tc2 cells. The umbilical cord blood MSCs has potential clinical application to prevention and treatment of GVHD.
