【摘要】： 目的:鉴定抗云锡矿工肺鳞癌单克隆抗体(C6)的特异性、纯化其抗体,为进一步获取YTMLC-90(云锡矿工肺鳞癌)肿瘤抗原蛋白及肿瘤抗原基因、探索其发生机制奠定基础。方法:采用免疫印迹(Western blot),检测各种肿瘤细胞、人正常细胞和各种肿瘤组织及正常组织与抗云锡矿工肺鳞癌单克隆抗体(C6)的交叉反应。采用亲和层析法纯化该抗体;用免疫印迹方法鉴定纯化单抗。结果:抗云锡矿工肺鳞癌单克隆抗体(C6)与各种肺癌细胞及肺癌组织抗原有交叉反应,而与其它肿瘤细胞、肿瘤组织及人正常细胞、正常组织抗原无交叉反应。纯化出了抗云锡矿工肺鳞癌单抗(C6)。结论:抗云锡矿工肺鳞癌单克隆抗体(C6)与肺癌相关抗原有较高的特异性,而与其它肿瘤相关抗原及正常组织抗原无特异性。获得有活性的纯化单抗。
[Abstract]:Objective: to identify the specificity of monoclonal antibody (C6) against lung squamous cell carcinoma (LSCC) of Yunnan tin miners and to purify its antibody in order to obtain tumor antigen protein and tumor antigen gene of YTMLC-90 (Yunxi miner lung squamous cell carcinoma). It lays a foundation for exploring the mechanism of its occurrence. Methods: Western blotters were used to detect various tumor cells. The cross-reaction of human normal cells, various tumor tissues and normal tissues with monoclonal antibody (C6) against Yunnan tin miners' lung squamous cell carcinoma was used to purify the antibody by affinity chromatography. The purified McAb was identified by Western blot. Results: the monoclonal antibody against lung squamous cell carcinoma (C6) was cross-reacted with various lung cancer cells and lung cancer tissue antigens, but with other tumor cells, tumor tissues and human normal cells. There was no cross reaction of normal tissue antigen. The monoclonal antibody against lung squamous cell carcinoma of Yuntin miners was purified. Conclusion: the monoclonal antibody (C6) against Yuntin miners' lung squamous cell carcinoma has high specificity with lung cancer associated antigens, but has no specificity with other tumor associated antigens and normal tissue antigens.
