神经前体细胞在成年小鼠脊髓的分布特征

【摘要】: 目的建立特异性表达巢蛋白(nestin)的转基因小鼠动物模型,观察和分析成年动物脊髓各个部位表达巢蛋白的阳性细胞数,探讨神经前体细胞(neural precursor cells ,NPCs)在脊髓各个部位的分布特征,为诱导自体神经前体分化修复脊髓损伤的研究提供更多的理论依据。 方法采用巢蛋白第二内含子增强子控制的LacZ(β-半乳糖苷酶)启动子转基因小鼠(nestin second-intron enhancer controlled LacZ reporter transgenic mice, pNes-Tg)和LacZ染色及阳性细胞量化技术,观察分析脊髓各个部位的巢蛋白表达阳性细胞的分布特征,即表达部位和表达数量。 结果神经前体细胞广泛分布在成年小鼠的整个脊髓的各个部位,但是在不同区域其数量和密度不同,在脊髓后角分布最多,其次是中央管,前角和侧角分布最少。在脊髓后角区神经前体细胞主要分布在胶状物质,在中央管神经前体细胞主要分布在室管膜带,在脊髓髓质(白质)区和神经核团分布很少,灰质比白质分布多,左右两边分布无明显差别。神经前体细胞在脊髓的各个节段中分布有显著差异,在颈段的分布最多,其次是腰骶段,在胸节段最少。但是同一节段(C1-7, T1-12, L1-5和骶段)的神经前体细胞分布没有显著差异。 结论在脊髓中神经前体细胞的分布特征可能与临床脊髓的损伤和修复有相关性,神经前体细胞分布较少的区域易于损伤且难于修复,像前角、胸节段,相反,在后角、中央管神经前体细胞分布最多,这些区域临床较少发生损伤,且较易修复。神经前体细胞在神经元分布区域分布较少,而在神经胶质细胞分布区域和室管膜带分布较多,这也提示神经前体细胞的衍化与神经胶质细胞和室管膜细胞有密切关系。
[Abstract]:Objective to establish a transgenic mouse model with specific expression of nestin (nestin), observe and analyze the number of positive cells expressing nestin in various parts of spinal cord of adult animals, and investigate the distribution characteristics of neural precursor cells (NPCs) in different parts of spinal cord of adult animals. To provide more theoretical basis for the study of inducing autologous neural precursor differentiation to repair spinal cord injury. Methods the nestin second-intron enhancer controlled LacZ reporter transgenic mice( pNes-Tg) and LacZ staining and positive cell quantification technique were used in the transgenic mice controlled by the second intron enhancer of nestin (尾 -galactosidase). The distribution of nestin positive cells in different parts of spinal cord was observed and analyzed. Results nerve precursor cells were widely distributed in all parts of the spinal cord of adult mice, but their numbers and densities were different in different regions, and most of them were in the posterior horn of spinal cord, followed by the central canal. The distribution of anterior horn and lateral horn was the least. In the posterior horn of spinal cord, nerve precursor cells were mainly distributed in colloid substance, in central canal nerve precursor cells were mainly distributed in ependymal zone, and in medullary matter (white matter) and nucleus of spinal cord. The distribution of nerve precursor cells in each segment of the spinal cord was significantly different, and the distribution in the cervical segment was the most, followed by the lumbosacral segment, the distribution of the gray matter was more than that of the white matter, and there was no significant difference between the left and right sides of the distribution. But there was no significant difference in the distribution of neural progenitor cells between C1-7, T1-12, L1-5 and sacral segments in the same segment. Conclusion the distribution of neural precursor cells in the spinal cord may be related to the injury and repair of the clinical spinal cord. Areas with less distribution of neural precursor cells are prone to injury and difficult to repair, such as the anterior horn, thoracic segment, and, on the contrary, in the posterior horn. The nerve precursor cells in the central duct were most distributed, which were less damaged in clinic and easier to repair. The neural progenitor cells were less distributed in the neuronal distribution area, but more in the glial cell distribution area and ependymal zone. This also suggests that the evolution of neural precursor cells is closely related to glial cells and ependymal cells.
