人卵巢癌高转移细胞株裸鼠原位移植瘤模型的建立

【摘要】: 背景与目的卵巢癌仍是病死率最高的妇科恶性肿瘤。理想的动物模型对于卵巢癌生物学行为及治疗方法的研究等方面均有重要作用。肿瘤原位模型因具有相对较高的仿真性而成为动物实验研究的首选。本研究选择人卵巢癌高转移细胞株HO-8910PM作为成瘤细胞,拟建立裸鼠卵巢癌原位移植瘤模型,并对相关生物学特性进行研究。 方法首先,将HO-8910PM细胞(约2×106个细胞)注射到雌性裸鼠皮下。待皮下成瘤后再通过显微外科手术将瘤体小块转移到其它裸鼠卵巢内,追踪观察原位肿瘤的生长及体内的转移情况,在荷瘤鼠处于濒死状态时将其处死,取原位及转移病灶处的肿瘤组织作光镜及电镜的病理检查。 结果共有14只裸鼠接受了原位移植手术,经追踪观察,有8只原位成瘤,成功率为57.14%。通过尸检发现有5只荷瘤鼠出现了远处器官的转移(转移率为83.33%),转移器官为肺,肝脏、膈肌、胃表面、肠管表面、胰脏、腹膜、对侧卵巢、髂窝淋巴结等处,以膈肌和肝脏较为普遍。 结论本研究成功地建立了裸鼠原位移植瘤模型,其生物学特性在某种程度上与人类卵巢癌更为相似,为卵巢肿瘤的研究提供了一种动物模型。
[Abstract]:Background & objective Ovarian cancer is still the most fatal gynecologic malignant tumor. The ideal animal model plays an important role in the study of biological behavior and treatment of ovarian cancer. In this study, the human ovarian cancer cell line HO-8910PM was selected as the tumor-forming cell. To establish an orthotopic transplanted tumor model of ovarian cancer in nude mice and to study its biological characteristics. Methods first, HO-8910PM cells (about 2 脳 106 cells) were injected subcutaneously into female nude mice. After subcutaneous tumorigenesis, the tumor was transferred to the ovary of other nude mice by microsurgery. The tumor growth and metastasis in vivo were observed, and the tumor tissues in situ and metastases were examined by light microscope and electron microscope when the tumor-bearing mice were on the verge of death. Results A total of 14 nude mice underwent orthotopic transplantation, 8 of them underwent orthotopic transplantation, and the success rate was 57.14.The distant organ metastasis was found in 5 tumor-bearing mice by autopsy (the metastasis rate was 83.33), and the metastatic organs were lung and liver. Diaphragm, stomach surface, intestinal canal surface, pancreas, peritoneum, contralateral ovary, iliac fossa lymph nodes are more common in diaphragm and liver. Conclusion the model of orthotopic transplanted tumor in nude mice was successfully established in this study, and its biological characteristics were more similar to those of human ovarian cancer to some extent, which provided an animal model for the study of ovarian tumor.
