基于生物信息学方法的H1N1流感病毒致病及传播特性研究

【摘要】: 流感病毒是一种造成人类、哺乳动物及鸟类患流行性感冒的RNA病毒,其传染性强、传播速度快,可引起急性呼吸道感染及多种并发症,严重威胁人类健康。通过生物信息学方法研究H1N1流感病毒致病及传播特性,有助于在基因水平上预防和治疗此类疾病。 本研究以H1N1流感病毒的HA、NA、NP、PA、PB2基因片段和人编码的microRNA为研究对象,选取2007年H1N1流感病毒20株、2008年H1N1流感病毒20株、2009年H1N1流感病毒20株、以及人编码的microRNA904条,结合序列比对和设计的microRNA筛选算法,预测人编码的调控H1N1流感病毒的microRNA,并将预测出的microRNA用于H1N1流感病毒的靶基因预测。通过靶基因预测结果的变化规律分析H1N1流感病毒的致病及传播特性。 本论文分为六个部分:第一部分,为绪论,简要介绍了本文的选题背景及研究的主要工作;第二部分,从理论上介绍了H1N1流感病毒的研究现状、生物信息学概况以及用生物信息学研究H1N1流感病毒的主要方法;第三部分,详细介绍了microRNA及其靶基因的研究方法;第四部分,详细叙述了人编码的调控H1N1流感病毒的microRNA预测方法;第五部分,通过预测出的调控H1N1流感病毒的microRNA,进一步预测其在H1N1流感病毒的靶基因;第六部分,论文的结果分析与展望,对研究过程中可能存在的问题和有待进一步研究的内容做了简要说明。 本研究说明,人编码的microRNA对于H1N1流感病毒具有调控能力。预测出的每条microRNA只作用于流感病毒的一个基因片段,且大部分microRNA只作用于H1N1流感病毒的同一个靶基因,也有少数microRNA作用于流感病毒一个基因片段的多个靶基因。人编码的microRNA对病毒调控能力的变化可能导致H1N1流感病毒致病及传播特性的变化。
[Abstract]:Influenza virus is a kind of RNA virus that causes influenza in humans, mammals and birds. It is highly contagious and spreads quickly. It can cause acute respiratory infection and many complications. The study on the pathogenicity and transmission of H1N1 influenza virus by bioinformatics is helpful to the prevention and treatment of this disease at the gene level. In this study, we selected 20 strains of H1N1 influenza virus in 2007, 20 strains of H1N1 virus in 2008, 20 strains of H1N1 influenza virus in 2009, and 20 human encoded microRNA904 fragments of H1N1 influenza virus. Combined with sequence alignment and designed microRNA filtering algorithm, The predicted microRNA was used to predict the target gene of H1N1 influenza virus. The variation of target gene prediction results were used to analyze the pathogenicity and transmission characteristics of H1N1 influenza virus. This thesis is divided into six parts: the first part, as the introduction, briefly introduces the background of the topic and the main research work, the second part, introduces the current research situation of H1N1 influenza virus in theory. The general situation of bioinformatics and the main methods of using bioinformatics to study H1N1 influenza virus. The third part introduces the research methods of microRNA and its target genes in detail. In this paper, the prediction method of microRNA coding for regulating H1N1 influenza virus is described in detail; in the fifth part, the target genes of H1N1 influenza virus are further predicted by the predicted microRNAs to regulate H1N1 influenza virus; in the sixth part, the results of the paper are analyzed and prospected. The possible problems in the research process and the contents to be further studied are briefly explained. This study shows that the human encoded microRNA has the ability to regulate the H1N1 influenza virus. Each predicted microRNA acts on only one fragment of the influenza virus, and most of the microRNA only acts on the same target gene of the H1N1 influenza virus. There are also a few microRNA acting on multiple target genes of a single gene fragment of the influenza virus. The changes in the regulatory ability of the human encoded microRNA to the virus may lead to changes in the pathogenicity and transmission characteristics of the H1N1 influenza virus.
