中国小肠结肠炎耶尔森菌分子流行病学研究

【摘要】: 小肠结肠炎耶尔森菌是一种重要的全球分布的人兽共患肠道致病菌,在人、多种动物、食品及外环境中都有分布。为阐明中国小肠结肠炎耶尔森菌分布特征,本研究对我国1980~2008年从腹泻病人、宿主动物、食品及环境分离到的1295株小肠结肠炎耶尔森菌,进行流行病学分布特征与血清型、毒力基因、脉冲场凝胶电泳(PFGE)分型等病原学特征的分析。结果表明,中国小肠结肠炎耶尔森菌地域分布广泛、宿主多样、血清型众多。而致病性小肠结肠炎耶尔森菌目前则仅存在于0:3和0:9两个血清型,且多分布于北方寒冷地区,但在少数温暖的沿海地区,0:3血清型致病性菌株也有分布。在同一地区内,从腹泻病人分离到的致病性菌株与部分动物源性菌株的PFGE型别一致,提示家畜家禽是人类感染小肠结肠炎耶尔森菌的重要来源,而猪是造成我国人类感染的最主要宿主,同时也证实农家犬也是引起我国部分地区人群感染的传染源。 本研究还第一次对小肠结肠炎耶尔森菌进行了多位点序列分析(MLST),阐释小肠结肠炎耶尔森菌MLST型别特征,并探索小肠结肠炎耶尔森菌与假结核耶尔森菌、鼠疫耶尔森菌的亲缘关系。结果表明,小肠结肠炎耶尔森菌与假结核耶尔森菌在MLST分型中分别属于两个不同类,鼠疫耶尔森菌则属于假结核耶尔森菌的一个分支。小肠结肠炎耶尔森菌MLST分析明显的形成致病性菌株与非致病性菌株两大类别。致病性菌株ST型别集中,围绕血清型呈现出高度的克隆性;而非致病性菌株型别分散,没有明显的流行病学关联,呈现明显的多态性。
[Abstract]:Yersinia enterocolitica (Yersinia enterocolitica) is an important global distribution of zoonotic enteropathogenic bacteria, distributed in human, animal, food and external environment, in order to elucidate the distribution characteristics of Yersinia enterocolitica in China, in order to elucidate the distribution of Yersinia enterocolitica in China. In this study, 1295 strains of Yersinia enterocolitica isolated from diarrhea patients, host animals, food and environment in China from 1980 to 2008 were studied. The epidemiological distribution, serotype and virulence genes of Yersinia enterocolitica were studied. The results showed that Yersinia enterocolitica was widely distributed in China, and its host was diverse. However, Yersinia enterocolitica is only present at 0:3 and 0:9, and most of the serotypes are distributed in cold areas of northern China. But in a few warm coastal areas, serotypic pathogenic strains were also distributed. In the same region, the PFGE types of pathogenic strains isolated from diarrhea patients were the same as those of some animal-derived strains. It is suggested that livestock and poultry are important sources of Yersinia enterocolitica infection in humans, and pigs are the main hosts of human infection in China, and it is also confirmed that farm dogs are also the source of infection in some parts of China. The multilocus sequence analysis of Yersinia enterocolitica was carried out for the first time to elucidate the characteristics of Yersinia enterocolitica MLST and to explore Yersinia enterocolitica and Yersinia pseudotuberculosis. The results showed that Yersinia enterocolitica and Yersinia pseudotuberculosis belong to two different classes in MLST typing. Yersinia pestis belongs to a branch of Yersinia pseudotuberculosis. The MLST analysis of Yersinia enterocolitica shows that there are two main types of pathogenic and non-pathogenic strains. The serotypes were highly clonal and non-pathogenic strains were dispersed and had no obvious epidemiological association and showed obvious polymorphism.
