野百合碱诱导大鼠肺动脉高压模型的实验研究

【摘要】:摘要目的:采用野百合碱(monocrotaline, MCT)诱导建立SD大鼠肺动脉高压模型,探讨野百合碱诱导肺动脉高压模型的稳定性及量效与时效关系,寻求野百合碱诱导肺动脉高压模型的最佳时间和剂量。方法:清洁级SD雄性大鼠110只,随机分为C组、M1、M2、M3和M4组,每组各15只,H1、H2和H3组,每组各12只。其中C组为对照组,给予相同剂量溶剂腹腔注射。M1~M4组大鼠分别一次性腹腔注射MCT30、40、50和60mg/kg,4周后比较各组大鼠的生存率、检测平均肺动脉压力(mPAP),肺动脉收缩压(PASP),右心室收缩压(RVSP),右心肥大指数(RVH1),并取肺组织行苏木素-伊红染色,观察肺的病理改变,进行肺小动脉形态计量学指标检测并通过肺微血管密度(MVD)的测定来综合判断肺动脉高压模型的稳定性。而H1~H3组分别一次性腹腔注射MCT50mg/kg,分别于2周、4周、6周后检测以上指标。结果:C组无死亡,M1~M4组大鼠的生存率分别为87%,87%,67%,40%。M1~M4各组SD大鼠平均肺动脉压、肺动脉收缩压、右心肥大指数依次增大(p<0.05),肺小动脉形态计量学指标检测显示肺血管中膜厚度百分比依次增大(p<0.05),肺微血管密度依次减小(p<0.05),而M3与M4组测量结果无明显差异。H2和H3组各项指标较H1组有明显差异,而H2和H3两组之间各项指标无明显差异。结论:腹腔注射50mg/kg剂量野百合碱4周后可诱导稳定肺动脉高压模型,是野百合碱诱导肺动脉高压的最佳剂量与时间。
[Abstract]:Objective: to establish a model of pulmonary hypertension in SD rats induced by monocrotaline (MCTT), and to investigate the stability, dose-effect and time-effect of monocrotaline induced pulmonary hypertension in SD rats.To find the best time and dose of monocrotaline induced pulmonary hypertension model.Methods: 110 male SD rats of clean grade were randomly divided into two groups: group C (n = 15) and group M _ 4 (n = 15) with H1H _ 2 and H _ 3, with 12 rats in each group.Group C was used as control group. The survival rate of rats in group C was compared after 4 weeks of intraperitoneal injection of MCT30 + 4050 and 60mg / kg respectively.The mean pulmonary artery pressure (MPP), pulmonary systolic pressure (PAP), right ventricular systolic pressure (RV), right cardiac fat index (RVH1) were measured. The lung tissues were stained with hematoxylin and eosin to observe the pathological changes of the lung.The stability of pulmonary hypertension model was evaluated by the measurement of pulmonary arterioles morphometry and the determination of pulmonary microvessel density (MVD).In H1~H3 group, 50 mg / kg MCT was injected intraperitoneally, and the above indexes were detected after 2 weeks and 4 weeks and 6 weeks.Results the survival rate of M1M4 rats without death was 87 and 87, respectively. The mean pulmonary artery pressure and pulmonary systolic pressure of SD rats in M1M4 group were significantly higher than those in control group 40. M1 and M4 groups, and the mean pulmonary arterial pressure and pulmonary systolic pressure in each group were significantly higher than those in control group (P < 0. 05).The right heart fat index increased in turn (P < 0.05), the pulmonary arterioles morphometrics showed that the percentage of pulmonary vascular media thickness increased in turn, and the pulmonary microvessel density decreased in turn, but there was no significant difference between M3 group and M4 group.There were significant differences between H1 group and H1 group.There was no significant difference between H _ 2 and H _ 3 groups.Conclusion: the stable pulmonary hypertension model can be induced by intraperitoneal injection of monocrotaline for 4 weeks, which is the best dose and time for monocrotaline to induce pulmonary hypertension.
