多巴胺对肾3型钠氢交换体的调控作用

【摘要】: 小剂量多巴胺具有利钠、利尿作用,并被广泛用于各种相关病症的治疗,但近年来多巴胺的作用逐渐受到质疑。由于肾脏局部可产生多巴胺,本实验给予低剂量左旋多巴,在无外周血管作用同时,研究其在肾脏局部对水盐代谢的调节作用。本实验通过RT-PCR?及?Western?blotting等方法分析了近曲小管NHE3和钠钾?ATP酶的表达及定位,结果发现:低剂量左旋多巴持续灌注并不影响血压及肾小球滤过率,但肾脏排水排钠增加。肾脏局部多巴胺水平增加使近曲小管内皮细胞?NHE3?表达降低,钠钾?ATP?酶水平未发生变化。由此推测多巴胺通过瞬时调节?NHE3快迅内化,从而减少钠的重吸收。综合各种检测结果,本研究认为肾脏局部多巴胺可通过诱导近曲小管细胞膜NHE3向胞内转移发挥利钠作用。多巴胺的利钠作用主要通过瞬时调节NHE3穿梭来实现的而不是通过长调节来实现的。近曲小管上皮细胞钠的重吸收?NHE3?起到限速作用,钠钾?ATP?酶只是钠重吸收的附助条件。
[Abstract]:Low dose dopamine has natriuretic and diuretic effects, and has been widely used in the treatment of various related diseases, but the role of dopamine has gradually been questioned in recent years.Since dopamine can be produced locally in the kidney, low dose levodopa was given to study the regulation of water and salt metabolism in the kidney without peripheral vascular effect.In this experiment, RT-PCR?The expression and localization of NHE3 and Na + K + ATPase in proximal convoluted tubules were analyzed by Western blotting. The results showed that low dose levodopa infusion did not affect blood pressure and glomerular filtration rate, but increased renal sodium drainage.The increase of local dopamine level in the kidney causes NHE3 to be expressed in the endothelial cells of proximal convoluted tubules.Decreased expression, Na + K ATP?The enzyme level did not change.It is inferred that dopamine can reduce sodium reabsorption by transient regulation of NHE3 internalization.In this study, we concluded that local dopamine could induce the transfer of NHE3 to the cell membrane of proximal convoluted tubules.The effect of dopamine sodium is mainly achieved by transient regulation of NHE3 shuttle rather than long regulation.Sodium reabsorption in epithelial cells of proximal convoluted tubulesPlay a role in limiting the speed, sodium potassium ATP?Enzyme is only a secondary condition for sodium reabsorption.
