铁皮石斛多糖对冠心病模型家兔心功能及心肌收缩能力的影响

【摘要】:目的:观察铁皮石斛多糖对冠心病模型家兔心功能、心肌收缩能力的影响,并探讨其相关作用机制。方法:30只健康家兔随机分为对照组、模型组、铁皮石斛多糖低、中、高剂量组5组,每组6只。采用左冠状动脉放置缩窄环方法,制备冠心病模型。低、中、高剂量组铁皮石斛多糖按100,200,400 mg·kg-1ig给予,每天1次,连续21 d。对照组、模型组ig给予等体积生理盐水。记录左室收缩压(LVSP),左室舒张末压(LVEDP),左室压力变化最大上升和下降速率(±dp/dtmax),动脉收缩压(BPs)和动脉舒张压(BPd),心室质量指数,RT-PCR法检测心肌细胞α-肌球蛋白(α-MHC)mRNA,β-肌球蛋白(β-MHC)mRNA表达,免疫组化检测肌动蛋白(actin)表达。结果:与对照组比较,模型组LVEDP,LVWI,RVWI均升高(P0.01,P0.05),而LVSP,±dp/dtmax,BPs均降低(P0.01,P0.05),α-MHC mRNA表达显著下降(P0.01),β-MHC mRNA升高(P0.05),模型组actin蛋白表达呈阳性的心肌细胞较多。与模型组比较,铁皮石斛多糖低、中剂量组LVEDP降低(P0.01,P0.05),而LVSP,±dp/dtmax,BPs均升高(P0.01,P0.05),α-MHC mRNA表达显著升高(P0.01),β-MHC mRNA表达下降(P0.05),各剂量组actin蛋白表达呈阳性的心肌细胞较少。结论:铁皮石斛多糖一定程度上恢复心肌舒缩能力、改善心功能。
[Abstract]:Aim: to investigate the effects of Dendrobium candidum polysaccharides on cardiac function and myocardial contractility in rabbits with coronary heart disease (CHD). Methods 30 healthy rabbits were randomly divided into control group, model group, Dendrobium candidum polysaccharide low, medium and high dose groups, with 6 in each group. Coronary artery disease (CHD) model was established by placing constrictive ring in left coronary artery. Dendrobium candidum polysaccharides in low, medium and high dose groups were given 100200400 mg kg-1ig once a day for 21 days. The control group and the model group were given the same volume of saline intravenously. Left ventricular systolic pressure (LVSPN), left ventricular end-diastolic pressure (LVEDPN), left ventricular pressure maximum rise and fall rate (卤DP / dtmax) and arterial diastolic pressure (DBP) were recorded. The expression of 伪 -MHCmRNAs (尾 -MHCmRNAs, 尾 -MHCN mRNA) in cardiomyocytes was detected by RT-PCR. The expression of actin was detected by immunohistochemistry. Results: compared with the control group, the RVWI of LVEDPN LVWIWI in the model group was significantly higher than that in the control group, while the expression of actin protein in the model group was significantly lower than that in the control group. The expression of 伪 -MHC mRNA was significantly decreased (P 0.01) and the expression of 尾 -MHC mRNA was higher in the model group than in the control group. The positive expression of actin protein in the model group was higher than that in the model group. The expression of actin protein in the model group was more than that in the model group. Compared with the model group, Dendrobium candidum polysaccharide was lower in the middle dose group than that in the middle dose group. However, the LVEDP in the middle dose group was lower than that in the control group, while that in the 卤DP / dtmaxus BP was higher than that in the model group. The expression of 伪 -MHC mRNA was significantly higher than that in the model group, and the expression of 尾 -MHC mRNA was decreased in the cardiac myocytes with positive expression of actin protein. Conclusion: Dendrobium candidum polysaccharides can improve cardiac function and improve cardiac function.
