【摘要】: 目的:探讨骨髓基质细胞在传代培养与诱导分化为神经元过程中Hes1和Mash1基因的表达变化及其作用。方法:采用全骨髓培养法体外分离培养获得骨髓基质细胞,倒置显微镜下观察其形态变化,应用半定量RT-PCR技术分析Hes1和Mash1基因在传代和诱导分化为神经元过程中的动态时序表达。结果:传至3代以后的骨髓基质细胞形态较为均一,诱导后细胞形态出现神经元样改变。在BMSCs传代过程中,Hes1的表达逐渐下降;Mash1在传至第5代时高表达,随后急剧下降。Hes1在P3的骨髓基质细胞诱导后第1天表达下降;Mash1在P3的骨髓基质细胞诱导后表达逐日增加。结论:Hes1和Mash1基因在骨髓基质细胞传代与诱导分化为神经元过程中可能起重要作用。 目的:研究大鼠脊髓全横断损伤后,Wnt信号基因在不同时相脊髓的表达变化及对内源性神经干细胞的作用。 方法:成年健康Wistar大鼠30只,随机分为假手术组(Sham组)和脊髓损伤组(SCI组),SCI组分为术后1、3、7、14、21 d组,每组5只。SCI组大鼠T11脊髓段做全横断损伤,Sham组仅行椎板切除术,术后大鼠进行BBB运动功能评分。采用半定量RT-PCR方法检测脊髓损伤组织中Wnt信号基因的表达。 结果:SCI组所有动物在术后均表现为双后肢瘫痪,BBB运动功能评分明显低于Sham组。SCI组在脊髓横断术后1、3、7 d Wnt3a表达均较Sham组明显增加;尾侧端Wnt3a较同时相头侧端的表达多,且在21 d又出现明显的升高。在Sham组和SCI组均未检测到Ngn1的表达。Hes1的表达在SCI后第1 d较Sham组有明显的下降,第3 d出现升高,之后逐渐下降。结论:Wnt信号可能参与了脊髓损伤后再生修复的过程。
[Abstract]:Aim: to investigate the expression and role of Hes1 and Mash1 genes in bone marrow stromal cells (BMSCs) during subculture and differentiation into neurons. Methods: bone marrow stromal cells were isolated and cultured in vitro by whole bone marrow culture method. The morphologic changes of bone marrow stromal cells were observed under inverted microscope. Semi-quantitative RT-PCR technique was used to analyze the dynamic temporal expression of Hes1 and Mash1 genes during passage and differentiation into neurons. Results: the morphology of bone marrow stromal cells was uniform after 3 passage, and neuronal changes appeared after induction. During the passage of BMSCs, the expression of Hes1 decreased gradually and the expression of Mash1 increased gradually at the fifth passage, and then decreased sharply. The expression of Mash1 in P3 bone marrow stromal cells decreased on the first day after induction. The expression of Mash1 in P3 bone marrow stromal cells increased day after day. Conclusion the gene of 1: Hes1 and Mash1 may play an important role in the passage and differentiation of bone marrow stromal cells into neurons. Aim: to study the changes of Wnt signal gene expression in different phases of spinal cord and its effect on endogenous neural stem cells (NSCs) after total transection of spinal cord in rats. Methods: thirty adult healthy Wistar rats were randomly divided into sham-operated group (Sham group) and spinal cord injury group (sci group). BBB motor function score was performed in rats after operation. Semi-quantitative RT-PCR was used to detect the expression of Wnt signal gene in spinal cord injury tissues. Results the results showed that the motor function score of all the animals in group 1: sci was significantly lower than that in group Sham.
