慢性阻塞性肺病寒饮蕴肺证大鼠模型肺组织生长因子的动态演变

【摘要】:目的:根据前期自然基金课题的研究,对慢性阻塞性肺疾病(chronicobstructive pulmonary disease,COPD)及寒饮蕴肺证进行理论探索和系统总结,,以肺组织生长因子VEGF、TGF-β1的动态演变为主要观测对象,探讨COPD寒饮蕴肺证病机动态演变与肺组织生长因子VEGF、TGF-β1的关系,以丰富和发展COPD寒饮蕴肺证理论。此课题延续采用寒凉刺激法结合烟熏及气管内滴注内毒素(LPS)以建立COPD寒饮蕴肺证大鼠模型,通过观察模型大鼠60天自然演变过程以及自拟温阳化饮方,布地奈德干预下肺组织生长因子VEGF、TGF-β1病机演变,来阐释COPD寒饮蕴肺证模型大鼠的肺组织生长因子VEGF、TGF-β1、肺功能、肺组织形态学的动态演变,及自拟温阳化饮方对COPD寒饮蕴肺证的模型大鼠肺组织生长因子VEGF、TGF-β1干预作用,来确定自拟温阳化饮方的作用性和时效性,为此病证防治提供新的思路和理论支撑。方法:运用烟熏结合气管内滴注LPS,饮用冰水(0℃冰箱中冷冻成冰水混合物)复制成COPD寒饮蕴肺证病证大鼠模型,观察(其中间隔10天,共60天)该模型的自然状态下的病机动态演变,并运用自拟温阳化饮方、布地奈德对该病证模型进行动态干预,并进一步观察该模型的一般情况,肺组织学,检测相关肺功能指标,肺组织生长因子VEGF、TGF-β1。结果:此实验是在前课题的基础上加以改进后复制而成COPD寒饮蕴肺证的大鼠模型,该模型的形态及行为、指标变化与COPD寒饮蕴肺证临床表现相符。模型大鼠吸气阻力、呼气阻力升高,肺顺应性下降,肺组织生长因子VEGF表达降低,TGF-β1表达升高,同时病理切片示肺组织出现了炎性损伤和纤维化变。在去除寒凉刺激因素后60天内,模型大鼠的症状、各项指标没有明显好转,并且有出现恶化的趋势,在40天左右大鼠寒饮蕴肺证开始弱化,有可能转变为其他证型。用自拟温阳化饮方进行干预的模型大鼠症状出现好转,肺功能有所改善,肺组织生长因子VEGF表达升高,TGF-β1表达降低,病理切片显示炎性损伤减轻,在40天左右达到峰值,基本接近正常大鼠水平。用布地奈德进行干预的模型大鼠症状也有所好转,但不能接近正常大鼠水平,其肺组织生长因子VEGF表达升高不明显,TGF-β1也无明显降低,病理切片能显示炎性损伤减轻。结论:运用寒凉刺激加烟熏且气管内滴注LPS造模的COPD寒饮蕴肺证大鼠模型是符合临床试验动物模型,具有良好的可靠性和稳定性。肺组织血管的改变对COPD寒饮蕴肺证影响颇大,肺组织生长因子VEGF表达降低,TGF-β1表达升高与肺组织血管的形成关系密切。运用自拟温阳化饮方干预能上调肺组织中生长因子VEGF的表达和下调TGF-β1的表达,对COPD寒饮蕴肺证疗效肯定,且对该病证的治疗上有时效性,肺组织生长因子VEGF、TGF-β1是其作用靶点之一。
[Abstract]:Objective: to explore and systematically summarize the theory of chronic obstructive pulmonary disease (pulmonary) and the syndrome of cold drink retention of the lung according to the research of natural fund, and to observe the dynamic evolution of lung tissue growth factor (VEGF) TGF- 尾 1. To explore the relationship between the dynamic evolution of COPD cold drink syndrome and the lung tissue growth factor VEGF-尾 1 in order to enrich and develop the theory of COPD cold drink accumulation lung syndrome. This subject continued to establish the rat model of COPD cold drink accumulation of lung syndrome by using the cold stimulation method combined with smoking and intratracheal instillation of endotoxin. By observing the natural evolution process of the model rats in 60 days and the self-made warming Yanghua decoction. The pathogenesis of lung tissue growth factor (VEGF) TGF- 尾 1 was induced by budesonide to elucidate the dynamic evolution of lung tissue growth factor (VEGF) TGF- 尾 1, lung function and lung tissue morphology in rats with COPD cold drink syndrome. And the intervention effect of WYYY decoction on lung tissue growth factor (VEGF) TGF- 尾 1 in model rats with COPD cold drink syndrome, to determine the function and timeliness of WYY decoction, and to provide new ideas and theoretical support for the prevention and treatment of the disease syndrome.
