慢性间歇性缺氧大鼠心肌氧化应激的研究

【摘要】: 目的: 制作可应用于常压慢性间歇性缺氧研究的动物模型,为进一步研究OSAHS (Obstructive sleep apnea-hypopnea syndrome)的生理病理提供一个实验平台。通过研究正常和慢性间歇性缺氧以及去除缺氧后大鼠心肌还原型谷胱甘肽(GSH)和蛋白质羰基化(PC)的变化来探讨OSAHS患者心肌是否存在氧化应激(OS)及氧化应激引起的进一步损害。 方法: 选取24只SD健康成年雄性大鼠,随机分为空白组(unhandled control, UC组)、慢性间歇性缺氧组(chronic intermittent hypoxia,CIH组)和去除缺氧组(Removal of hypoxia, RH组),UC组正常饲养,CIH组每日间歇缺氧8小时,连续4周,建立慢性间歇性缺氧模型,RH组前4周同CIH组,后4周正常饲养,所有大鼠在实验结束后取心肌0.5g再提取心肌蛋白进行免疫印迹来分析还原型谷胱甘肽(GSH)和蛋白质羰基化(PC)的变化。 结果: ①GSH的表达CIH组较UC组和RH组明显降低(p0.05),而UC组和RH组之间无明显差异(p0.05)。 ②PC的结果CIH组较UC组和RH组明显升高(p0.05),而UC组和RH组之间无明显差异(p0.05)。 结论: 慢性间歇性缺氧致使大鼠心肌存在氧化应激状态,并引起心肌蛋白的损伤。
[Abstract]:Purpose :



To establish an animal model applicable to the study of chronic intermittent hypoxia in normal pressure , and to provide an experimental platform for further study of the physiological and pathological features of obstructive sleep apnea - syndrome syndrome . By studying the changes of normal and chronic intermittent hypoxia and the changes of glutathione ( GSH ) and protein carbonylation ( PC ) in rats after hypoxia , it is possible to study the presence of oxidative stress ( OS ) and oxidative stress in the myocardium of patients with obstructive sleep apnea .



Method :



24 SD healthy adult male rats were randomly divided into blank group ( UC group ) , chronic intermittent hypoxia group ( group C ) , chronic intermittent hypoxia group ( CIH group ) and hypoxia group ( removal of hypoxia , RH group ) .



Results :



The expression of GSH increased significantly in CIH group than in UC group and RH group ( p < 0.05 ) , but there was no significant difference between UC group and RH group ( p < 0.05 ) .



The results of PC showed that the CIH group was significantly higher than that in UC group and RH group ( p < 0.05 ) , but there was no significant difference between UC group and RH group ( p < 0.05 ) .



Conclusion :



Chronic intermittent hypoxia causes oxidative stress in the myocardium of rats and causes damage to myocardial proteins .
