心肌缺血再灌损伤大鼠血清HSP25水平的动态变化及临床意义初探

【摘要】: HSP25(heat shock protein 25)是小分子量热休克蛋白之一,在细胞内具有保护细胞骨架、抗损伤、抗凋亡等多种生物学功能。近年来研究发现HSP27也可以释放到细胞外。有研究表明与健康人相比,急性冠状动脉综合症病人血浆中HSP27的水平大量增加,推测其可能是一个新的心肌损伤标志物。本研究探讨心肌缺血再灌损伤时大鼠血清HSP25(与HSP27同源)的动态变化及临床意义。 本研究首先采用结扎心脏冠状动脉左前降支的方法制备大鼠在体心肌缺血再灌注模型,在心肌缺血30min再灌注1h、2h、4h、6h、12h、24h从心脏处取血,检测血清HSP25、LDH、CK、CK-MB、Mb、cTnI的水平,观察心肌缺血再灌时血清HSP25、LDH、CK、CK-MB、Mb、cTnI水平的动态变化。其次用40℃1h、40℃6h、43℃30min HS处理H9C2大鼠心肌细胞株,观察细胞的LDH释放率及上清液中HSP25浓度的变化。结果显示:(1)再灌损伤组(I/R组)大鼠血清HSP25的水平从再灌注1h升高,2h达到峰值,4、6、12和24h逐渐回落,但仍高于正常水平。(2)大鼠心肌缺血再灌注损伤时,血清HSP25水平与LDH、CK、CK-MB、Mb水平具有正相关性。(3)40℃6h、43℃30minHS处理H9C2细胞株时,细胞LDH释放率增加,上清液中HSP25的浓度升高,与对照组相比有显著差异。上述结果提示:1.大鼠心肌缺血再灌注损伤时,小分子HSP25可以从受损心肌释放入血,导致血清HSP25的水平升高。2.鉴于心肌缺血再灌注早期血清HSP25的水平上升快,敏感性高,因此血清HSP25有作为心肌损伤早期诊断指标的潜力。
[Abstract]:HSP25 (heat shock protein 25) is one of the small molecular weight heat shock proteins. It has many biological functions such as protecting cytoskeleton, anti injury, anti apoptosis and so on. In recent years, it has been found that HSP27 can also be released to the cells. Compared with healthy people, the level of HSP27 in patients with acute coronary artery syndrome is greatly increased. In addition, it is presumed that it may be a new marker of myocardial injury. This study is to explore the dynamic changes and clinical significance of HSP25 (and HSP27 homologous) in the serum of rats with myocardial ischemia reperfusion injury.
In this study, a rat model of myocardial ischemia reperfusion was made by ligating the left anterior descending branch of the coronary artery. The blood of 1H, 2h, 4h, 6h, 12h, 24h was taken from the heart after myocardial ischemia 30min, and the serum HSP25, LDH, CK, CK-MB, Mb, and the serum levels were detected. Then, the myocardial cells of H9C2 rats were treated with 40 1H, 40 6h and 43 C 30min HS. The LDH release rate and the HSP25 concentration in the supernatant were observed. The results showed that: (1) the level of HSP25 in the serum of the reperfusion injury group (I/R group) increased from the reperfusion 1H, 2h reached the peak, and was gradually falling down, but still higher than the normal level. (2) The serum HSP25 level was positively correlated with the level of LDH, CK, CK-MB and Mb in the rat myocardial ischemia reperfusion injury. (3) at 40 C, 6h, and 30minHS treatment at 43 C H9C2 cell lines, the LDH release rate increased and the concentration of HSP25 in the supernatant increased significantly compared with the control group. The results suggested that the scores of myocardial ischemia reperfusion injury in 1. rats were small. HSP25 can be released into the blood from damaged myocardium, causing the level of serum HSP25 to rise in.2., in view of the rapid rise of serum HSP25 level and high sensitivity in the early stage of myocardial ischemia reperfusion, so serum HSP25 has the potential as an early diagnostic indicator of myocardial injury.
