双侧坐骨神经切断对猫脊神经节蛋白质组表达变化的实验研究

【摘要】: [目的]检测猫双侧坐骨神经切断后所属脊神经节蛋白质组的变化。 [方法]健康成年雄性猫4只,切断双侧坐骨神经,动物存活3天后取材,分别提取脊神经节组织总蛋白,测定其浓度。采用双向电泳技术纯化分离出蛋白质;应用2-DE图谱分析软件PDQuest,分析出各组间双向电泳图谱中表达差异的蛋白点;选取明显差异蛋白点做基质辅助激光解析电离飞行时间质谱和串联质谱分析,获取肽指纹图谱和肽序列标签,用Mascot软件从NCBI数据库中检索确定蛋白质名称。 [结果]获得重复性较好的双向电泳图谱,经质谱鉴定,坐骨神经切断脊神经节组与正常对照组间存在4个差异蛋白:神经微丝重多肽、热休克蛋白90β、γ烯醇酶、3-磷酸甘油醛脱氢酶。此四种蛋白在坐骨神经切断后的所属脊神经节中缺失。 [结论]上述四个缺失蛋白质,可能参与了坐骨神经切断后引发的所属脊神经节的病理反应过程。
[Abstract]:[objective] to detect the proteome changes of spinal ganglion after bilateral sciatic nerve transection in cats. [methods] the sciatic nerve was cut off from 4 healthy adult male cats. The total protein of spinal ganglion was extracted and the concentration was measured after the animals survived for 3 days. The protein was purified and separated by two-dimensional electrophoresis, and the protein spots were analyzed by 2-DE analysis software. Obviously different protein spots were selected for matrix assisted laser desorption ionization time of flight mass spectrometry and tandem mass spectrometry to obtain peptide fingerprints and peptide sequence tags. The protein names were retrieved from NCBI database by Mascot software. [results] Two-dimensional electrophoretic patterns with good reproducibility were obtained. The results showed that there were four different proteins between the sciatic nerve transection spinal ganglion group and the normal control group: neural microfilament weight polypeptide. Heat shock protein 90 尾, 纬 -enolase 3-glyceraldehyde phosphate dehydrogenase. These four proteins are absent in the spinal ganglion of the sciatic nerve. [conclusion] the above four missing proteins may be involved in the pathological reaction of the spinal ganglion after sciatic nerve transection.
