变性I型胶原对晚代骨髓间充质细胞成骨能力的影响

【摘要】: 目的 探讨变性的Ⅰ型胶原(DC)作为培养基质对体外培养的骨髓间充质干细胞的增殖和成骨能力的影响。 方法 体外培养大鼠的骨髓间充质干细胞,分为A、B两组,A组在铺有变性的Ⅰ型胶原膜的培养皿中培养;B组在没有铺变性的Ⅰ型胶原膜的培养皿中培养。培养至第八代时,分别检测两组细胞的增殖活性和细胞调亡率;并对细胞进行成骨细胞诱导,进行碱性磷酸酶染色和定量分析。 结果 对于体外培养的第八代骨髓间充质干细胞,相比较普通培养的细胞,生长在变性的Ⅰ型胶原膜上的细胞表现出较高的增殖活性和ALP表达,并且有较少的细胞发生调亡。 结论 变性的Ⅰ型胶原作为骨髓间充质干细胞的体外培养基质,可减缓细胞由于多次传代而引起的细胞衰老,并可保持细胞的成骨分化能力。为组织工程提供了一种可获得大量有增殖活性和分化能力的种子细胞的途径。
