青春期酒精刺激对大鼠成年后空间学习记忆以及海马结构突触囊泡素表达的影响

【摘要】: 目的:探讨青春期酒精刺激对大鼠成年后学习记忆与海马结构突触囊泡素表达的影响 方法:取青春期40只SD大鼠分为对照组与实验组,两组动物分别以生理盐水、2g/kg酒精连续注射5天,每天固定时间2次,然后正常饲养,待成年后用Morris水迷宫对各组大鼠进行空间辨别性学习记忆的测试,用摄像机拍摄大鼠的行为,再用统计分析软件分析水迷宫检测结果。用免疫组织化学方法检测正常对照组与实验组大鼠海马结构突触囊泡素的表达。 结果:Morris水迷宫结果:各组大鼠随着训练次数的增加逃避潜伏期(EL)所用时间逐渐缩短。酒精组逃避潜伏期(EL)比正常对照组长,差异有统计学意义(P<0.05)。 突触囊泡素免疫表达:突触囊泡素的免疫表达主要集中表达在海马结构的CA_3以及齿状回,为褐色颗粒状,呈板状分布。正常对照组与酒精组比较,大鼠海马结构CA_3跟齿状回突触囊泡素免疫反应阳性产物的平均灰度值差异有统计学意义(P<0.05)。 结论:青春期接受酒精刺激的大鼠,成年后海马结构CA_3、齿状回突触囊泡素表达减少,Morris水迷宫测试中的逃避潜伏期延长,空间学习与记忆能力下降。
[Abstract]:Objective: to investigate the effects of alcohol stimulation during adolescence on learning and memory and synaptic vesicles expression in hippocampus of adult rats. Methods: forty puberty SD rats were divided into two groups: control group and experimental group. The animals in the two groups were injected with saline 2g / kg alcohol for 5 days, fixed twice a day, then fed normally. The spatial discriminant learning and memory were tested by Morris water maze after adulthood. The behavior of rats was photographed by video camera and the results of water maze detection were analyzed by statistical analysis software. The expression of synaptophysin in hippocampal structure of normal control group and experimental group was detected by immunohistochemical method. Results: the time of escaping latency (EL) of rats in each group was shortened with the increase of training times. The escape latency (EL) of alcohol group was longer than that of normal control group (P < 0. 05). Immunoreactivity of synaptophysin: the immunoreactivity of synaptophysin was mainly expressed in CA_3 and dentate gyrus of hippocampal structure. Compared with alcohol group, the average gray value of CA_3 and synaptic vesicle immunoreactive products in rat hippocampal structure was significantly different from that in alcohol group (P < 0. 05). Conclusion: the expression of synaptic vesicles in CA_3, dentate gyrus of adult rats exposed to alcohol in adolescence decreased the escape latency prolonged and the ability of spatial learning and memory decreased in Morris water maze test.
