云南白族男性人群Y-SNP遗传多态性研究

【摘要】:[目的]本课题基于Y染色体系统进化树,选择O、C、N支上的43个Y-SNP位点,采用MassARRAY技术对云南大理白族男性人群Y-SNP位点进行调查研究,获得云南大理白族男性人群单倍群分布频率以及遗传结构状况,建立Y-SNP基础DNA数据库,为法医学应用奠定基础。 [方法]在知情同意的原则下,严格按照准入标准(男性无关个体,三代以内均为白族,无血缘关系),在云南大理州及其他地州采集278例云南白族男性个体的外周静脉血5ml, EDTA-Na2抗凝,采用MassARRAY法检测实验样本Y染色体上43个SNP位点的遗传多态性,应用SPSS19.0统计学软件进行统计学分析。 本课题采用MassARRAY技术,对样本进行逐级筛选,分析了O支P101、L682、 M175、M324、M134、2611、M50(M110)、P164、Page23(M117)、M164、M162_1、 Page20(M119)、P201、F706、M312、P31、Pk4、F238、M95、L127.1、 F11、M88、M159、M188(M113)、M122、M101、P203.1的位点,分析了C支的V20、M347、M38、M105、M216、M356、M210的位点,以及分析了N支的L666、M231、L729、P189.2、L708、L732、M46(P105)位点,共计43个SNPs位点进行筛选分析。 [结果]获得云南白族男性人群Y-SNP单倍型以及单倍型频率,在278个无关白族男性个体中,共检出28个Y-SNP单倍型,云南白族男性人群Y-SNP单倍群多样性为0.0642。其中M175、M324、M134、P164、P201在白族人群中的单倍群分布频率较高。白族未检出单倍群P101、L682、M50、M164、M162_1、M312、P49、 V20、M347、M38、M105、P1892、L732、M356、M210。对于43个Y-SNPs位点来说,白族对应的基因多样性范围从0.0000到0.6753。 [结论]依据人类Y染色体系统进化树,分析云南白族278例男性样本的43个Y-SNPs位点,获得28个Y-SNP单倍群、单倍群分布频率以及遗传结构信息,与文献报道的白族以及其他少数民族的Y-SNP单倍群分布频率进行分析,得出结论。不但可弄清中国云南大理白族的起源以及民族间的相互关系,研究各民族基因组结构差别,分析其差别的遗传学意义,为人类进化和中国各民族源流、迁徙和划分提供生物学依据,而且尝试建立用MassARRAY分析检测方法为云南人群Y-SNP研究提供相关数据,同时为建立云南基础DNA数据库提供数据。
[Abstract]:[objective] based on the phylogenetic tree of Y chromosome, 43 Y-SNP loci were selected from the N branch of Oligon, and MassARRAY technique was used to investigate the Y-SNP loci in the male population of Dali Bai nationality in Yunnan Province. The distribution frequency of haploidy and genetic structure of male population of Bai nationality in Dali Yunnan Province were obtained. The basic DNA database of Y-SNP was established to lay the foundation for forensic application. [methods] under the principle of informed consent, the admission criteria were strictly followed. In Dali Prefecture and other prefectures of Yunnan Province, the peripheral venous blood 5ml and EDTA-Na2 anticoagulant were collected from 278 male individuals of Yunnan Bai nationality.
