冻干人用狂犬病脂质体疫苗的研究

【摘要】: 人用狂犬病疫苗暴露前免疫程序一般为0、7、21或28天各接种1剂量的疫苗(2.5IU/剂),暴露后免疫为0、3、7、14、28天各接种1剂量的疫苗(2.5IU/剂)。该免疫程序的缺点是接种针次多,全程免疫时间长。 本文以脂质体作为狂犬病疫苗的佐剂,制备了高效价的冻干人用狂犬病脂质体疫苗(LipoRabV),目的是减少全程免疫针次,缩短全程免疫时间。本文以小鼠及比格犬为实验动物对LipoRabV进行了细胞免疫研究,体液免疫研究及疫苗保护效果研究。证明了脂质体对狂犬病疫苗的佐剂作用,提高了疫苗的保护效力。在细胞免疫研究中,冻干人用狂犬病脂质体疫苗促进了特异性T淋巴细胞的增殖,促进了细胞因子IL-2、IFN-γ的产生。体液免疫研究表明,LipoRabV 3针免疫产生的中和抗体不低于冻干人用狂犬病疫苗(RabV)5针免疫的中和抗体水平。在暴露后免疫程序研究中,LipoRabV 3针免疫保护率高于对照RabV5针免疫的保护率。 本文初步确定了冻干人用狂犬病脂质体疫苗的免疫程序:暴露前免疫程序为0、14天各接种1剂量的冻干人用狂犬病脂质体疫苗(6.0IU/剂);暴露后免疫程序为0、3、14天各接种1剂量的冻干人用狂犬病脂质体疫苗(6.0IU/剂)。同现有疫苗相比,暴露前免疫程序由3针减至2针,全程免疫由28天减至14天。暴露后免疫程序由5针减至3针,全程免疫由28天减至14天。 本项目于2006年被立为国家“863”项目,项目编号为2006AA02A221。
[Abstract]:The routine of human rabies vaccine immunization before exposure was 1 dose of 2.5IU/ on 21 or 28 days after exposure, and 1 dose of 2.5IU/ was inoculated with 1 dose of 2.5IU/ on each day after exposure. The drawback of the immune program is that the vaccination times are many and the whole immunization time is long. Using liposome as adjuvant of rabies vaccine, a freeze-dried human rabies liposome vaccine (LipoRabV),) with high titer was prepared in this paper. In this paper, the cellular immunity, humoral immunity and vaccine protection of LipoRabV were studied in mice and Beagle dogs. The effect of liposome on the adjuvant of rabies vaccine was proved, and the protective effect of the vaccine was improved. In the study of cellular immunity, freeze-dried human rabies liposome vaccine promoted the proliferation of specific T lymphocytes and the production of cytokine IL-2,IFN- 纬. Humoral immunity studies showed that the neutralizing antibody produced by LipoRabV 3 doses was not lower than that of freeze-dried human vaccinated with rabies vaccine (RabV) 5 doses. In the study of post-exposure immune program, the protective rate of LipoRabV 3 was higher than that of control RabV5. The immunization procedure of lyophilized human rabies liposome vaccine was preliminarily determined: the immunization procedure before exposure was 0 ~ 14 days after exposure, 1 dose of freeze-dried human rabies liposome vaccine (6.0IU/) was inoculated before exposure, and after exposure, the procedure of immunization was 0 ~ 3 ~ 3 ~ 14 days each. 1 dose of lyophilized human rabies liposome vaccine (6.0IU/). Compared with the existing vaccine, the procedure of immunization before exposure was reduced from 3 to 2, and the whole immunization was reduced from 28 days to 14 days. After exposure, the immunization procedure was reduced from 5 to 3, and the whole immunization was reduced from 28 to 14 days.
