补肾活血法对大鼠慢性高眼压模型外侧膝状体损伤的干预及作用机制研究

【摘要】:目的:观察补肾活血法对大鼠慢性高眼压(elevated intraocular pressure, EIOP)模型外侧膝状体(lateral geniculate nucleus,LGN)损伤的干预作用,初步探讨补肾活血法对其的作用机理。 方法:采用烙闭上巩膜静脉法,烙闭大鼠3支上巩膜静脉,建立大鼠慢性EIOP模型,随机分为3组:空白组,模型组,给药组。连续灌胃8周,并于8周末处死大鼠,观察补肾活血法对EIOP大鼠眼压(intraocular pressure, IOP)、LGN病理形态学变化以及对脑源性神经营养因子(brain-derived neurotrophin factor, BDNF)表达的影响。 结果:本实验采用的烙闭上巩膜静脉的造模方法使大鼠IOP明显升高(P0.01),在造模后8周(实验结束)IOP仍为造模前的2.5-3倍;补肾活血法对大鼠慢性EIOP模型LGN损伤的干预作用如下:轻度降低IOP;修复EIOP大鼠对LGN的损伤;提高EIOP大鼠LGN的BDNF表达。 结论:采用烙闭上巩膜静脉的方法建立慢性EIOP大鼠模型,操作简单、效果稳定,可获得不少于两个月的中等度EIOP;补肾活血法可以有效的保护慢性EIOP大鼠模型视功能,其作用主要表现在轻度降低IOP,修复EIOP大鼠对LGN的损伤,提高EIOP大鼠LGN的BDNF表达。
[Abstract]:Objective: to observe the effect of Bushen Huoxue method on (lateral geniculate nucleus,LGN injury of lateral geniculate body of chronic high intraocular pressure (elevated intraocular pressure, EIOP) model in rats, and to explore the mechanism of Bushen Huoxue method. Methods: chronic EIOP model of rats was established by cauterization of superior scleral vein and three branches of superior scleral vein of rats. The rats were randomly divided into three groups: blank group, model group and administration group. The rats were perfused continuously for 8 weeks and killed at the end of 8 weeks. The effects of Bushen Huoxue method on the pathological changes of intraocular pressure (intraocular pressure, IOP), LGN) and the expression of brain-derived neurotrophic factor (brain-derived neurotrophin factor, BDNF) in EIOP rats were observed. Results: the rat IOP was significantly increased by cauterization of scleral vein (P0.01), and the IOP was still 2.5-3 times as high as that before modeling at the end of the experiment at 8 weeks after the establishment of the model. The intervention effects of Bushen Huoxue method on LGN damage of chronic EIOP model in rats were as follows: mild decrease of LGN damage in EIOP rats repaired with IOP; and increase BDNF expression of LGN in EIOP rats. Conclusion: chronic EIOP rat model is established by cauterization of the superior scleral vein. The operation is simple, the effect is stable, and the moderate EIOP; of not less than two months can be obtained. Bushen Huoxue method can effectively protect the visual function of chronic EIOP rat model. The main effect of this method is to reduce the LGN damage caused by IOP, repair in EIOP rats and to increase the BDNF expression of LGN in EIOP rats.
