转基因大鼠构建技术和转SV40Tag大鼠脑肿瘤模型建立的研究

【摘要】: 转基因技术对推动现代生物学和医学发展产生了巨大的作用。转基因小鼠技术已经成为了一种常规的实验室技术,目前已经建立了数千种不同的转基因小鼠模型,为生物医学研究提供了有效的在体模型工具。但转基因大鼠的研究在国内却是进展缓慢,转基因效率很低,为了提高转基因大鼠的效率,本研究对不同组合PMSG和HCG对不同周龄大鼠超排后胚胎数量、胚胎质量、原核注射后存活率和胚胎移植成功率等进行了系统分析,通过三种不同基因转基因大鼠模型构建成功建立了原核注射转基因大鼠技术。在此基础上我们又将pBC-SV40Tag注射到SD大鼠胚胎原核中,经PCR和Southern检测成功获得了SV40Tag阳性转基因大鼠。RT-PCR及westhern bloting结果表明:pBC-SV40Tag转基因大鼠不仅如预期的在胰腺组织表达SV40Tag,同时也在脑中高表达SV40Tag,并成功获得了脑肿瘤转基因大鼠模型。
[Abstract]:Transgenic technology plays an important role in promoting the development of modern biology and medicine. Transgenic mouse technology has become a conventional laboratory technology, and thousands of different transgenic mouse models have been established, which provides an effective in vivo model tool for biomedical research. In order to improve the efficiency of transgenic rats, the number and quality of superovulation embryos in different combinations of PMSG and HCG were studied. The survival rate after prokaryotic injection and the success rate of embryo transfer were systematically analyzed. The technique of prokaryotic injection transgenic rats was successfully established through the construction of three transgenic rat models with different genes. On this basis, we injected pBC-SV40Tag into the prokaryotic cells of SD rat embryos. SV40Tag positive transgenic rats were successfully obtained by PCR and Southern. The results of RT-PCR and westhern bloting showed that pBC-SV40Tag transgenic rats expressed SV40Tag, not only in pancreas as expected, but also in brain. The transgenic rat model of brain tumor was successfully obtained.
