两种氨基葡萄糖对兔膝骨性关节炎模型关节液中IL-1和TNF-α含量影响的实验研究

【摘要】:目的:本实验通过检测盐酸/硫酸氨基葡萄糖治疗兔膝骨关节炎时关节液中IL-1和TNF-α含量来探讨盐酸/硫酸氨基葡萄糖(glucosamine hydrochloride,OTL/glucosamine sulfate,WGL)对骨性关节炎的作用及机制。 方法:选取健康雄性成年大白兔36只,体重2.0-2.5kg。随机分为3组(n=12):对照组(A组)、盐酸氨基葡萄糖组(B组)、硫酸氨基葡萄糖组(C组)。三组均按改良Hulth法制备膝骨性关节炎模型;B、C组于造模术后第2开始每天分别灌胃盐酸/硫酸氨基葡萄糖(90mg/kg),连续8周;A组自由饮食。伤口愈合后每天驱赶所有兔子行走2次,30min/次。给药8周后拍摄对照组兔子双侧膝关节DR片,对比双侧膝关节影像学变化;空气栓塞法处死所有实验兔,收集右膝关节液,用ELISA法检测关节液中IL-1和TNF-α含量;解剖对照组兔双侧膝关节,观察双侧股骨髁肉眼改变并评分。 结果:实验动物全部存活,伤口愈合良好。对照组(A组)双侧膝关节DR片对比可见右膝关节周软组织肿胀较明显,内侧关节间隙狭窄,呈膝骨性关节炎改变。肉眼观A组右股骨髁软骨面剥脱、缺损严重,可见软骨下骨质外露,以股骨内侧髁为重;A组左侧股骨髁软骨面光滑、色泽正常;双侧膝关节软骨评分有统计学差异(P0.05)。三组关节液中均可检测出IL-1和TNF-α,与A组比较,B组与C组关节液中IL-1和TNF-α均下降,有统计学意义(P0.05),B组与C组比较无明显差异(P0.05)。 结论:改良Hulth造模方法可以制备出膝骨性关节炎模型;盐酸氨基葡萄糖与硫酸氨基葡萄糖可能通过抑制关节液中炎性因子IL-1和TNF-α的表达,延缓了软骨病变的速度,盐酸氨基葡萄糖与硫酸氨基葡萄糖无明显差异。
[Abstract]:Objective: to investigate the content of IL-1 and TNF- 伪 in articular fluid of rabbit knee osteoarthritis treated with hydrochloric acid / glucosamine sulfate (glucosamine hydrochloride,OTL/glucosamine sulfate,). The effect and mechanism of WGL on osteoarthritis. Methods: 36 healthy male adult white rabbits were selected, weighing 2.0-2.5 kg. They were randomly divided into 3 groups: control group (group A), glucosamine hydrochloride group (group B) and glucosamine sulfate group (group C). The knee osteoarthritis model was established by modified Hulth method in all the three groups, the rats in group C were given hydrochloric acid / glucosamine sulfate (90mg/kg) daily for 8 weeks, group A was fed with free diet. After wound healing, all rabbits were driven to walk twice a day, 30min/. After 8 weeks of administration, the DR film of bilateral knee joint of the control group was taken and the imaging changes of bilateral knee joint were compared. All the experimental rabbits were killed by air embolization, and the contents of IL-1 and TNF- 伪 in the joint fluid were detected by ELISA method, and the right knee joint fluid was collected. The bilateral knee joint of the control group was dissected and the changes of bilateral femoral condyle were observed and scored. Results: all the experimental animals survived and the wound healed well. In the control group (group A), the soft tissue swelling around the right knee joint was obvious, the medial joint space was narrow, and the knee osteoarthritis was observed on DR film of bilateral knee joint. In group A, the right femoral condyle cartilage was dissected, the defect was serious, the subchondral bone was exposed, the medial femoral condyle was the most serious, the left femoral condylar cartilage in group A was smooth and the color was normal. Bilateral knee cartilage score was statistically different (P0.05). IL-1 and TNF- 伪 were detected in all three groups. Compared with group A, IL-1 and TNF- 伪 in group B and group C decreased significantly (P0.05). There was no significant difference between group), B and group C (P0.05). Conclusion: the model of knee osteoarthritis can be made by modified Hulth method. Glucosamine hydrochloride and glucosamine sulfate may inhibit the expression of inflammatory factors IL-1 and TNF- 伪 in articular fluid, and delay the speed of cartilage lesion. There is no significant difference between glucosamine hydrochloride and glucosamine sulfate.
