ALS基因mRNA在白念珠菌不同生物状态的表达

【摘要】: 目的通过对酵母状态和芽管状态的白念珠菌ALS基因mRNA表达水平的比较,以及对浮游状态和生物膜状态的白念珠菌ALS基因mRNA表达水平的比较,探讨白念珠菌ALS基因mRNA在白念珠菌芽管及生物膜形成中可能发挥的作用,为防治白念珠菌感染提供重要的理论依据。 方法选取4株标准株白念珠菌和11株临床株白念珠菌进行实验研究。白念珠菌酵母状态的培育和芽管状态的诱导以及浮游状态和生物膜状态白念珠菌的制备:GSB培养基,25℃振荡培养48h,收获酵母状态的白念珠菌;ph7.0的RPMI-1640液体培养基,37℃振荡培养3h,收获芽管状态的白念珠菌;体外在6孔细胞培养板上,RPMI-1640液体培养基,37℃静置培养48h,收获生物膜状态的白念珠菌。热酸性酚法抽取不同生长状态白念珠菌的总RNA,RNA的纯度经紫外分光光度计A260/A280比值估计,RT-PCR两步法逆转录ALS2基因mRNA、ALS3基因mRNA以及ACT1基因mRNA,cDNA扩增,扩增产物琼脂糖凝胶电泳,凝胶成像系统拍照。利用Quantity One电泳分析软件对ALS2基因和ALS3基因的cDNA扩增条带进行分析,应用SPSS11.5医学统计软件包的计数资料配对样本差值均数的t检验对实验数据进行分析(以P<0.05为显著性标准)。 结果标准株白念珠菌芽管状态的ALS2基因mRNA(P=0.005,P<0.01)和ALS3基因mRNA(P=0.0022,P<0.01)相对表达水平均明显高于酵母状态,临床株白念珠菌芽管状态的ALS2基因mRNA(P=0.000,P<0.01)和ALS3基因mRNA(P=0.000,P<0.01)相对表达水平均明显高于酵母状态,其中标准株和临床株白念珠菌中多数菌株未检测ALS3基因mRNA的表达,个别菌株酵母状态ALS3基因mRNA仅检测到低水平的表达,;标准株白念珠菌生物膜状态的ALS2基因mRNA(P=0.0276.P<0.05)和ALS3基因mRNA(P=0.0405,P<0.05)相对表达水平均明显高于浮游状态;临床株白念珠菌生物膜状态的ALS2基因mRNA(P=0.0004,P<0.01)和ALS3基因mRNA(P=0.0005,P<0.01)相对表达水平均明显高于浮游状态。 结论ALS2基因mRNA表达和ALS3基因mRNA表达可能在白念珠菌丛酵母相向菌丝相的转换过程中起重要作用。ALS2基因mRNA和ALS3基因mRNA的表达在体外白念珠菌生物膜形成中可能起着一定作用。
