CGRP对HaCaT细胞增殖和凋亡及分泌NGF的影响

【摘要】: 目的:本文研究降钙素基因相关肽(CGRP)对HaCaT细胞增殖、凋亡及分泌NGF的影响。 方法:以不同浓度(10~(-10)mol/L~10~(-7)mol/L)CGRP作用于HaCaT细胞,用MTT法检测对细胞增殖及生长曲线的影响;流式细胞术分析对细胞凋亡的影响;酶联免疫吸附测定(ELISA)法检测细胞上清液中NGF浓度的变化。应用SPSS 13.0统计软件包进行统计学处理,差别有显著性标准为P<0.05。 结果:CGRP促HaCaT细胞增殖作用的强度与CGRP的浓度和作用时间有关:CGRP浓度为10~(-10)mol/L至10~(-8)mol/L时,其促增殖作用呈浓度依赖性(F=7.824 P<0.01);当浓度上升至10~(-7)mol/L时促增殖作用较10~(-8)mol/L时减弱(F=15.231 P<0.01);CGRP作用48小时促增殖作用最强;流式细胞术分析发现随CGRP浓度升高,其抑制HaCaT细胞凋亡的作用增强;ELISA检测发现HaCaT细胞上清液中NGF浓度随CGRP浓度升高和作用后时间延长而增加(F=32.207、76.520 P<0.01)。 结论:CGRP可促进HaCaT细胞增殖、抑制其凋亡,并可促进HaCaT细胞分泌NGF。HaCaT细胞分泌NGF增加可能是CGRP促进增殖、抑制凋亡的原因之一。
[Abstract]:Aim: to study the effects of calcitonin gene-related peptide (CGRP) on the proliferation, apoptosis and secretion of NGF in HaCaT cells. Methods: HaCaT cells were treated with different concentrations (10 ~ (-10) mol/L~10~ (-7) mol/L) CGRP, and the effects on cell proliferation and growth curve were detected by MTT assay, and apoptosis was analyzed by flow cytometry. Enzyme linked immunosorbent assay (ELISA) was used to detect the concentration of NGF in supernatant. SPSS 13.0 statistical software package was used for statistical analysis, and the difference was significant (P < 0.05). Results: the effect of CGRP on the proliferation of HaCaT cells was related to the concentration and time of CGRP. When the concentration of CGRP was from 10 ~ (-10) mol/L to 10 ~ (-8) mol/L, the effect of CGRP on proliferation of HaCaT cells was concentration-dependent (FF7. 824 P < 0. 01). When the concentration increased to 10 ~ (-7) mol/L, the effect of promoting proliferation was weaker than that of 10 ~ (-8) mol/L (F _ (15.231) P < 0. 01); CGRP). Flow cytometry showed that the inhibitory effect of CGRP on apoptosis of HaCaT cells was enhanced with the increase of CGRP concentration, and that the concentration of NGF in the supernatant of HaCaT cells increased with the increase of CGRP concentration and the prolongation of the time after treatment by ELISA assay (FN 32.207? 76.520 P < 0. 01). Conclusion: CGRP can promote the proliferation and inhibit the apoptosis of HaCaT cells and increase the secretion of NGF by NGF.HaCaT cells of HaCaT cells which may be one of the reasons of CGRP promoting proliferation and inhibiting apoptosis.
