人巨细胞病毒UL146基因-α趋化因子在巨细胞病毒感染中作用的研究

【摘要】: 人巨细胞病毒UL 146基因—α趋化因子在巨细胞病毒感染中作用的研究 目的 研究人巨细胞病毒(HCMV)UL146基因在巨细胞病毒感染临床株中的多态性,寻找具有临床诊断、治疗价值的基因及基因的变化,为HCMV感染的临床诊断和防治提供资料。 方法 选择经QPCR检测尿HCMV-DNA为阳性的临床株进行UL146基因序列PCR扩增,对扩增阳性标本进行序列测定和分析。 结果 28株扩增阳性标本序列之间有很大变异,种系树化分析结果显示可以分为3个型,4个亚型,G1组氨基序列与Toledo株的同源性为96.6%-98.3%,G2组为10.3%-22.2%,G3组为12.8%-13.7%;ELRCXC的趋化因子功能区在各序列中高度保守。 结论 HCMV-UL146基因呈现高度多态性,其编码的ELRCXC趋化因子功能区在各序列中高度保守,HCMV-UL146基因多态性与HCMV感染不同临床症状之间无明显的关系。
[Abstract]:Study on the role of human cytomegalovirus UL 146 gene 伪 chemokine in cytomegalovirus infection objective to study the polymorphism of human cytomegalovirus (HCMV) UL146 gene in clinical cytomegalovirus infection. To search for the gene and gene changes with clinical diagnosis and therapeutic value, and to provide data for clinical diagnosis and prevention and treatment of HCMV infection. Methods the UL146 gene sequence was amplified by PCR from clinical strains which were positive for urinary HCMV-DNA detected by QPCR. The sequence of positive samples was determined and analyzed. Results there was great variation among 28 amplified positive specimens. The results of phylogenetic analysis showed that there were 3 types and 4 subtypes. The amino sequence of G1 group was homologous to Toledo strain, the homology was 96.6- 98.3%. In G2 group, 10.3- 22.2and G3 were 12.8- 13.7; The chemokine functional regions of ELRCXC are highly conserved in each sequence. Conclusion HCMV-UL146 gene is highly polymorphic and its ELRCXC chemokine functional region is highly conserved in each sequence. There is no significant relationship between HCMV-UL146 gene polymorphism and different clinical symptoms of HCMV infection.
