【摘要】: 目的: 1、研究睡眠剥夺对大鼠血浆中去甲肾上腺素(NE)、多巴胺(DA)、五羟色胺(5-HT)含量及认知功能的影响。 2、探讨NE、DA、5-HT与睡眠—觉醒周期的内在联系。 3、探讨睡眠剥夺影响认知功能的可能机制。 方法: 健康雄性8~12周龄的Sprague-Dawley(SD)大鼠27只,通过随机数字表分为三组:(1)小平台睡眠剥夺组;(2)大平台环境对照组;(3)空白对照组。采用改良多平台睡眠剥夺法建立睡眠剥夺模型,睡眠剥夺前后用Morris水迷宫视频跟踪分析系统检测三组大鼠的认知功能水平。采用腹主动脉取血法收集血浆,沉淀蛋白质,利用高效液相电化学法检测血浆中NE、DA、5-HT的含量。 结果: 1、血浆中NE、DA、5-HT的检测结果:(1)NE:睡眠剥夺组的含量比环境对照组、空白对照组的含量高,差异有统计学意义(p值分别为0.033、0.034),环境对照组与空白对照组的含量无明显差异。(2)DA:睡眠剥夺组的含量较环境对照组、空白对照组的含量低,差异有统计学意义(p值分别为0.034、0.035),环境对照组与空白对照组的含量无明显差异。(3)5-HT:睡眠剥夺组的含量较环境对照组、空白对照组的含量高,差异有统计学意义(p值分别为0.036、0.035),环境对照组与空白对照组的含量无明显差异。 2、Morris水迷宫检测大鼠认知功能的结果:(1):睡眠剥夺前,睡眠剥夺组、环境对照组和空白对照组的潜伏期无统计学差异。(2):睡眠剥夺后,睡眠剥夺组、环境对照组和空白对照组三组的潜伏期均下降,差异有统计学意义(p值分别为0.047、0.045、0.043)。(3):睡眠剥夺后,睡眠剥夺组的潜伏期比环境对照组、空白对照组的潜伏期明显较长,差异有统计学意义(p值分别为0.044、0.043),环境对照组和空白对照组的潜伏期无统计学差异。 结论: 1、睡眠剥夺影响体内神经递质的含量,使血浆中NE、5-HT水平明显升高,DA水平下降。 2、睡眠剥夺使机体的认知功能下降。 3、体内NE、DA、5-HT含量的改变可能与睡眠—觉醒周期存在一定的联系。 4、睡眠剥夺对认知功能的影响可能与体内NE、DA、5-HT含量的改变相关。
[Abstract]:Objective: 1. To study the effects of sleep deprivation on the contents of norepinephrine, dopamine, serotonin (5-HT) and cognitive function in plasma of rats. 2. To explore the inner relationship between NEDAA 5 HT and sleep-wakefulness cycle. 3. To explore the possible mechanism of sleep deprivation affecting cognitive function. Methods: Twenty-seven healthy male Sprague-Dawley (SD) rats aged 8 to 12 weeks were randomly divided into three groups: 1) small platform sleep deprivation group; 2) large platform environment control group; 3) blank control group. Sleep deprivation model was established by modified multi-platform sleep deprivation method. The cognitive function of the three groups was measured by Morris water maze video tracking analysis system before and after sleep deprivation. Plasma was collected and protein was precipitated by abdominal aorta blood sampling. A high performance liquid-phase electrochemical method was used to determine the content of neodadine 5-HT in plasma. Results: The results showed that the content of NED 5-HT in plasma was higher in the sleep deprivation group than in the environmental control group and the blank control group. The difference was statistically significant (P = 0.033, 0.034). There was no significant difference between the environmental control group and the blank control group. The content of sleep deprivation group was higher than that of the environment control group. The content of blank control group was low, the difference was statistically significant (0.034% 0.035). There was no significant difference between the environmental control group and the blank control group. The content of sleep deprivation group was higher than that of the environment control group, and the content of blank control group was higher. The difference was statistically significant (P = 0.036). There was no significant difference between the environmental control group and the blank control group. 2 the results of Morris water maze test of cognitive function in rats: 1: 1: before sleep deprivation, sleep deprivation group. There was no significant difference in latency between the environmental control group and the blank control group: after sleep deprivation, the latency of the sleep deprivation group, the environment control group and the blank control group were all decreased. The difference was statistically significant: the latency of sleep deprivation group was higher than that of environment control group after sleep deprivation. The incubation period of the blank control group was longer than that of the control group, and the difference was statistically significant (0.044 卤0.043), while the latency of the environmental control group and the blank control group had no statistical difference. Conclusion: 1. Sleep deprivation affected the content of neurotransmitters in vivo and significantly increased the level of NE-5-HT in plasma and decreased DA level. 2. Sleep deprivation reduces the cognitive function of the body. 3. The change of NED 5-HT may be related to the sleep-wakefulness cycle. 4. The effect of sleep deprivation on cognitive function may be related to the change of NED 5 HT content.
