生殖管道阻塞对调节性T细胞的影响

【摘要】:目的:本实验通过结扎小鼠一侧或双侧输精管,造成输精管梗阻,精子输出受阻,光镜观察睾丸、附睾的变化,检测小鼠外周血CD4+CD25+调节性T细胞,探讨输精管梗阻小鼠睾丸的形态学变化以及梗阻对CD4+CD25+调节性T细胞的影响。方法:选用健康成年雄性BALB/c小鼠,随机分为3组,每组30只小鼠:对照组(A组,麻醉后仅进行阴囊切开后缝合),单侧输精管结扎组(B组,麻醉后结扎左侧输精管),双侧输精管结扎组(C组,麻醉后两侧输精管均结扎)。分别手术后10周、20周、26周(每个时间点各组取10只小鼠)应用光镜观察石蜡包埋HE染色后的小鼠睾丸、附睾的组织结构,流式细胞术检测外周血CD4+CD25+CD127-调节性T细胞百分比。结果:1、HE染色观察:输精管结扎小鼠睾丸形态学变化主要表现为结扎10周组生精小管结构不清,生精细胞有减少。2、输精管结扎一段时间后,实验小鼠外周血CD4+CD25+CD127-调节性T细胞百分比较对照组明显增高。结论:随着输精管梗阻时间的延长,机体对精子产生了免疫耐受,CD4+CD25+CD127-调节性T细胞显著增加,参与诱导免疫耐受的产生。
[Abstract]:Objective: to observe the changes of testis and epididymis by ligating one or both vas deferens of mice, resulting in obstruction of vas deferens and blocked sperm output, and to detect the regulatory T cells of CD4 CD25 in peripheral blood of mice. To investigate the morphological changes of testis and the effect of obstruction on CD4 CD25 regulatory T cells in mice with vasectomy. Methods: healthy adult male BALB/c mice were randomly divided into 3 groups, 30 mice in each group: control group A; After anesthesia, only scrotal incision and suture were performed, unilateral vasectomy group (group B), left vasectomy group (group C) and bilateral vasectomy group (group C). After anesthesia, both vas deferens were ligated. The tissues of the testis and epididymis were observed by light microscope at 10 weeks and 20 weeks after operation (10 mice in each group were taken at each time point), and the tissues of the testis and epididymis were observed by light microscope after paraffin-embedded HE staining. The percentage of CD4 CD25 CD127- regulatory T cells in peripheral blood was detected by flow cytometry. Results the morphological changes of testis in vas deferens ligated mice were characterized by unclear structure of seminiferous tubules after 10 weeks of ligation. The percentage of CD4 CD25 CD127- regulatory T cells in peripheral blood of the experimental mice was significantly higher than that of the control group. Conclusion: with the prolongation of the obstructive time of the vas deferens, the percentage of CD127- regulatory T cells in peripheral blood of the experimental mice was significantly higher than that of the control group. Immune tolerance T cells (CD4 CD25 CD127- regulatory T cells) produced by organism to spermatozoa were significantly increased, which was involved in the induction of immune tolerance.
