青藤碱对大鼠类风湿关节炎动物模型模式识别受体TLR-2影响的实验研究

【摘要】:目的:已有的研究提示青藤碱可以抑制模式识别受体的下游信号分子表达和信号转导,但其机理尚无深入的研究。本课题提出了青藤碱通过影响模式识别受体治疗类风湿关节炎的假说,通过在RA动物模型器官组织和炎症介质水平上的研究,探索青藤碱对模式识别受体的影响与治疗RA的相关性,为青藤碱的上市后再评价提供实验室依据。 方法:雄性SD大鼠50只,随机分为5组,建立Ⅱ型胶原免疫大鼠诱导的关节炎模型(CIA)。以青藤碱高、中、低治疗组与空白组、模型组作对照,定期测量大鼠体重和大鼠踝关节肿胀度,对大鼠踝关节滑膜组织作病理形态学观察(HE染色),并取各组大鼠的关节滑膜组织,检测TLR-2、NF-κB及相关细胞因子(TNF-α、IL-1)的表达率,运用SPSS软件进行统计分析。 结果:1、类风湿关节炎大鼠模型体重增长速度减慢,青藤碱能够有效抑制这种现象:2、青藤碱能够有效的减轻大鼠踝关节的肿胀;3、青藤碱能够降低大鼠TLR-2的表达水平,抑制大鼠关节滑膜炎症相关指标NF-κB、TNF-α、IL-1的表达,抑制关节滑膜损伤、关节骨的破坏:4、随着给药剂量的增加,肿胀度越低,滑膜损伤和骨破坏越轻微,相关指标的表达量也随之降低:5、高剂量青藤碱组与中、低剂量组比较大鼠体重有所下降。 结论:青藤碱通过抑制APC细胞的模式识别受体TLR-2的表达,抑制下游NF-κB的激活,使NF-κB信号通道的传导受到抑制,阻断APC的活化,使IL-1、INF-α等细胞因子的生成和分泌减少,抑制滑膜细胞的活化,使炎症反应和骨破坏得到抑制和延缓,从而达到抗类风湿关节炎的作用。
