青浦区266株结核分枝杆菌基因分型及其与耐药表型关系研究

【摘要】:目的探讨上海市青浦区流行的结核分枝杆菌(MTB)的主要基因型及其与耐药表型的关系。方法分别提取266株MTB DNA,应用多重聚合酶链反应(PCR)对其进行分型(北京基因型/非北京基因型)鉴定;采用比例法对此266株菌株进行利福平、异烟肼、链霉素和乙胺丁醇的药物敏感性试验;运用SPSS 17.0统计软件分析基因分型与患者性别、年龄的关系,以及基因分型与菌株耐药性之间的关系。结果 266株菌株中北京基因型242株(90.98%),非北京基因型24株(9.02%)。北京基因型菌株的总耐药率为23.96%,耐多药率为9.91%,非北京基因型的总耐药率和耐多药率分别为29.16%和8.33%,两者差异无统计学意义(P均0.05)。结论北京基因型MTB可能是上海市青浦区的主要流行株,尚未发现北京基因型菌株和非北京基因型菌株在耐药方面有明显差异。
[Abstract]:Objective to investigate the main genotypes of Mycobacterium tuberculosis (MTB) and its relationship with drug resistance phenotype in Qingpu District, Shanghai. Methods 266 MTB DNA, strains were identified by multiplex polymerase chain reaction (PCR), and rifampicin and isoniazid were detected by proportional method. The drug sensitivity test of streptomycin and ethambutanol, the relationship between genotyping and patient sex, age, and the relationship between genotyping and drug resistance were analyzed by SPSS 17.0 software. Results among 266 strains, 242 (90.98%) were Beijing genotypes and 24 (9.02%) were non-Beijing genotypes. The total drug resistance rate of Beijing genotype was 23.96, the multi-drug resistance rate was 9.91, the total drug resistance rate and multi-drug resistance rate of non-Beijing genotype were 29.16% and 8.33%, respectively. There was no significant difference between them (P 0.05). Conclusion Beijing genotype MTB may be the main epidemic strain in Qingpu district of Shanghai, but there is no obvious difference between Beijing genotype strain and non-Beijing genotype strain in drug resistance.
