人脐带间充质干细胞向肝样细胞分化的体外研究

【摘要】: 目的探讨人脐带间充质干细胞分离、培养的方法,研究其基本生物学特性。初步探讨人脐带间充质干细胞向肝样细胞诱导分化的方法。 方法无菌条件下收集剖宫产新生儿脐带,利用PBS冲洗脐动脉及脐静脉,将血液冲洗尽,将其剪碎至1mm~3后,加入Ⅳ型胶原酶(0.1%)消化60min,然后加入胰酶(0.125%)消化30min,利用滤网过滤后收集细胞,DMEM(5%FBS)重悬细胞接种于培养瓶内进行原代培养,待细胞生长至80-90%融合后进行1:3传代培养,取传代培养细胞进行形态学观察、免疫细胞化学染色、流式细胞仪检测、生长活性、细胞周期分析、染色体数目分析及软琼脂克隆形成实验。进一步取第6代细胞经胰酶消化后接种于12孔板中,待细胞生长至约70%融合后,利用两步诱导法进行诱导,对照组用原培养液培养。诱导组中加入0.5μM/L地塞米松,50 g/L ITS,20μg/L HGF,10μg/L FGF4,烟碱0.61g/L,共10天。后期加入20μg/L OSM,0.5μM/L地塞米松,50 g/LITS。每3d换液1次,在不同诱导阶段收集细胞后观察细胞形态,免疫细胞化学,RT-PCR,糖原染色。 结果利用胶原酶及胰酶两步消化法能充分消化脐带,细胞易于获得,收集细胞后进行原代培养,24小时可观察到贴壁细胞,待生长至10天时可见细胞长满瓶底,进行传代培养,每周传代1次。观察细胞形态为长梭形,不规则形,细胞大小不一,类似成纤维细胞,呈漩涡样生长。取第6-9代细胞行免疫细胞化学染色结果显示细胞强表达CD105、Vimentin,基本不表达CD34、CD31、CD133。经流式细胞仪检测表达CD29、CD49、CD105,基本不表达造血细胞标志CD34。MTT实验显示细胞倍增时间为22h。细胞周期分析表明75%-85%细胞处于G_0/G_1。染色体分析可见细胞染色体条数正常。在软琼脂中无克隆形成。经诱导后的细胞在培养到第14d可见细胞形态出现变化,细胞长突起逐渐消失,逐渐转为圆形或卵圆形,经免疫组化检测可见细胞在第1周开始表达AFP,第2周时CK18、CK19、OV6的表达被观察到。在诱导第3周时ALB表达被观察到,AFP表达逐渐减弱。到第4周AFP的表达基本消失,ALB的表达增强。RT-PCR结果显示细胞于第1周开始表达AFP,至诱导后期,AFP表达水平逐渐降低。于第3周时观察到ALB表达,随时间延长ALB表达逐渐增强。于第4周糖原染色呈阳性结果。 结论:经酶消化法可从脐带中获得间充质干细胞,易于获得,体外增殖能力强,与骨髓来源间充质干细胞的免疫表型相一致,符合间充质干细胞基本的生物学特性,有望在干细胞的研究及临床应用方面发挥重要的作用。脐带间充质干细胞在本实验诱导条件下可转化为肝样细胞,表达肝细胞的表面标志,初步具备肝细胞特有的功能性特征,将为肝细胞组织工程提供理想的细胞来源。
